翼状胬肉组织中环氧化酶-2及p21-ras的表达研究

【摘要】 目的通过免疫组织化学方法检测翼状胬肉组织中环氧化酶-2(COX-2)和p21ras的表达,进一步证实氧自由基在翼状胬肉形成和发展中的作用。方法选择临床切除的静止期、进行期和复发期胬肉材料共40例,行苏木精-伊红染色及COX-2和p21ras免疫组织化学染色。结果COX-2及p21ras在正常球结膜中表达微弱,静止期胬肉组织中COX-2和p21ras阳性染色主要位于复层鳞状上皮化生细胞,在进行期及复发期翼状胬肉组织中COX-2及p21ras阳性染色弥漫性分布于复层鳞状上皮化生细胞的胞浆和杯状细胞的胞浆中,胬肉组织中的炎性细胞和血管内皮细胞胞浆可见少量COX-2及p21ras表达。结论p21ras and COX-2的过度表达在翼状胬肉的发生和发展过程中可能起重要作用。

【Abstract】 ObjectiveThe pathological characteristics of pterygium are thought to be superficial squamous metaplasia,global cell density increase,proliferation and degeneration of conjunctival strom.The goal of this study was to detect the expression of cyclooxgenase-2 (COX-2) and p21ras in pterygium by immunohistochemistry and determine the oxygen-derived free radicals effect on the pathologenesis of pterygium.MethodsForty specimens of pterygium were collected from patients with primary and recurrent pterygia during the surgery.The normal conjunctiva tissue from Shenzhen Eye Bank was as control.The expressions of COX-2 and p21ras were detected using immunohistochemial method.The microstructures of these specimens were observed under the light microscope.ResultsThe COX-2 and p21ras were faintly expressed in the basal epithelial cells in normal conjunctiva tissue.Stationary stage of pterygia tissue showed positive reaction for COX-2 and p21ras mainly at stratified spuamous epithelium,exhibiting a brown-yellow staining in cell nuclear(p21ras) and cytoplasm(COX-2) of metaplastic cells by immunochemistry.The expression of COX-2 and p21ras was stronger in the active stage and the recurrence pterygia tissues,blood vessel endothelial cell and inflammatory cell such as lymphocyte,plasma cell,little amount of neutrophilic granulocyte and mastocyte,displaying a deeply brown dye in cytoplasm of metaplastic cells and goblet cells of stratified spuamous epithelium.ConclusionThe overexpression of p21ras and COX-2 may play the important role on the growth of pterygium.
