前房相关免疫偏离对碱烧伤眼角膜移植的影响
Effect of anterior chamber-associated immune deviation on corneal allografts in burned eyes
【摘要】 目的观察诱导供体特异性的前房相关免疫偏离(ACAID)是否有助于角膜植片在碱烧伤高危眼中的存活。方法用1mol/L的NaOH烧灼新西兰白兔单眼角膜中央建立高危眼模型,然后将动物随机分为诱导组和对照组,1个月后进行角膜移植。诱导组在角膜移植前10d前房植入供体肌肉诱导供体特异性ACAID,对照组未处理。观察诱导组与对照组相比角膜植片存活时间是否延长。结果与对照组相比,诱导ACAID组角膜植片平均存活时间延长,两组植片存活率的差异具有显著统计学意义。结论诱导供体特异性ACAID有助于碱烧伤高危眼角膜植片的存活。
【Abstract】 ObjectiveA main cause of corneal allografts failure is donor-specific delayed type hypersensitivity(DTH),while anterior chamber-associated immune deviation(ACAID)can suppress antigen-specific DTH.The paper was to assess the effect of donor-specific ACAID induction on the survival of orthotopic corneal allografts in alkali burned high-risk eyes.MethodsCentral corneal alkali burn injuries were induced in New Zealand white rabbits by 1mol/L NaOH to establish single-eye high-risk model.Experimental rabbits were randomly allocated into induced group(10 rabbits)and control group(10 rabbits),and orthotopic keratoplasty was performed 1 month later in both groups.Donor-specific ACAID was induced by inoculating muscle tissue of donors in 4℃ PBS and putting it into recipients anterior chamber in induced group in 10 days prior to keratoplasty whereas the eyes from control rabbits was untreated.The survival time and the histological changes of allografts between two groups were compared.ResultsCorneal allografts rejection are determined by neovascularization,opacity or edema.Survival time of corneal allografts in the eyes of induced group was markedly enhanced (56.7±10.5 days) compared with untreated controls (32.0±3.4 days) (P<0.05).The more new blood vessels and inflammatory cells were found in allografts of control group,while those of induced group presented less neovessels and infiltration of inflammatory cells.ConclusionInduction of donor-specific ACAID can effectively prolongs survival time of corneal allograft and suppress the rejection of corneal allograft in alkali burned high-risk eyes.
