人卵丘细胞线粒体膜电位与卵母细胞成熟度相关关系的研究
The relationship between human mitochondrial membrane potential of cumulus cells and oocyte maturation
【摘要】 目的:通过检测人类卵母细胞周围卵丘细胞的线粒体膜电位(△Ψm),研究其与卵母细胞成熟度的关系。方法:收集卵母细胞质内单精子注射(ICSI)的166个卵母细胞周围的卵丘细胞,根据卵母细胞的成熟度及受精情况分为3组:不成熟卵母细胞组23例;成熟卵母细胞未受精组18例;成熟卵母细胞受精组103例。分别测定线粒体膜电位。用透射电镜观察3例未成熟卵母细胞、3例成熟卵母细胞周围卵丘细胞线粒体的变化。结果:未成熟卵母细胞组、成熟卵母细胞未受精组、成熟卵母细胞受精组线粒体膜电位分别为1.5±0.43,3.9±1.68,5.8±2.24,两两比较差异有统计学意义。透射电镜观察发现未成熟卵母细胞周围卵丘细胞质内线粒体肿胀、嵴减少、嵴消失、空泡化的发生率较成熟卵母细胞周围卵丘细胞高。结论:卵丘细胞线粒体膜电位与相应卵母细胞的成熟度相关。
【Abstract】 Objective:To analyze the relationship between mitochondrial membrane po- tential(△Ψm)of cumulus cells and oocyte maturation.Method:Cumulus cells were obtained from oocytes undergoing intracytoplasmic sperm injections.According to the maturity of oocytes and fertilization,the oocytes were divided into three groups:the immature,the matured but un- fertilized,the matured and fertilized.The change of mitochondrial membrane potential was ob- served by transmission electron microscopy.Result:Significant differences of△Ψamong the three groups were identified.There was more damage rate of mitochondrial in immature oocytes than in matured oocytes.Conclusion:The change of△Ψm in human cumulus cells may reflect the maturation of oocyte.
