5-氟尿嘧啶纳米磁性颗粒在荷瘤鼠体内靶向性分布
Targeting Distribution of 5-FU Magnetic Nanoparticles in Nude Mice Bearing Hepatoma
【摘要】 目的研究5-氟尿嘧啶(5-FU)纳米磁性颗粒在荷瘤裸鼠体内的靶向性分布。方法制备荷瘤小鼠异位肝癌模型(荷瘤鼠),随机分3组,每组8只。A组:5-FU原液治疗组;B组:单纯5-FU纳米磁性颗粒治疗组,无磁场应用;C组:实验组,在肿瘤内部建立300高斯(GS)的磁场,药物应用同B组。采用高效液相色谱(HPLC)分析法检测离体组织中5-FU药物浓度,原子吸收光谱法(AAS)测定荷瘤鼠组织中的铁浓度,磁共振成像(MRI)检测5-FU纳米磁性颗粒在荷瘤鼠体内组织分布。结果在肿瘤组织内建立内磁场后,尾静脉注射5-FU纳米磁性颗粒(250 mg/kg),与无磁场的相同药物治疗组及单纯5-FU对照组比较,HPLC检测荷瘤鼠的肿瘤组织中5-FU浓度显著增加(P<0.01),AAS检测肿瘤组织中铁浓度也显著增加,肿瘤组织MRI的T2加权像发生变化,呈现低信号。结论在磁场引导下,5-FU纳米磁性颗粒中磁性载体和所载药物在荷瘤鼠体内具有肿瘤靶向性分布。
【Abstract】 Objective To evaluate the targeting distribution of 5-FU magnetic nanoparticles in nude mice bearing hepatoma.Methods Twenty-four nude mice bearing ectopic hepatoma were divided equally into 3 groups on random:group A receiving 5-FU administered via tail vein injection at 25 mg/kg on consecutive 5 days;group B receiving 5-FU magnetic nanoparticles administered via tail vein injection at 250 mg/kg on consecutive five days without applying magnetic fields;group C receiving both magnetic field and 5-FU magnetic nanoparticles.High performance liquid chromatography(HPLC) method for the determination of 5-FU in tissues was established and applied to determine 5-FU in mouse tissue samples.A flame atomic absorption spectrometer(AAS) was used to detect the iron concentration in mouse tissues.MRI was used to detect the distribution of targeting magnetic nanoparticles in vivo.Results The concentrations of both 5-FU and Fe in tumor tissues were increased significantly in group C as compared with those in group A and group B, and the signal intensity of tumor tissue in group C was weaker than in groups A and B.Conclusion As an integrative medicine,5-FU magnetic nanoparticle possesses targeting distribution in the treatment of nude mice bearing hepatoma based on the induction of magnetic field.
