硒对氟致L-02细胞DNA损伤和凋亡的影响
Effects of Selenium on DNA Damage and Apoptosis of L-02 Cell Induced by Fluoride
【摘要】 目的探讨硒对氟导致的L-02细胞DNA损伤和凋亡的影响。方法采用80μg/ml氟化钠和/或1.73μg/ml亚硒酸钠对培养的L-02细胞进行染毒,24 h后采用单细胞凝胶电泳技术(SCGE)检测各组L-02细胞DNA损伤情况;用流式细胞术(FCM)检测各组L-02细胞凋亡率。结果氟组L-02细胞DNA损伤率和凋亡率明显高于对照组、硒组和氟硒组(P<0.01);而对照组L-02细胞DNA损伤率和凋亡率与硒组和氟硒组之间无显著性差异(P>0.05)。结论氟可导致L-02细胞DNA损伤,诱导细胞凋亡,适量硒可拮抗氟导致的L-02细胞DNA损伤和细胞凋亡作用。
【Abstract】 Objective To explore the effects of selenium on DNA damage and apoptosis of L-02 cell induced by fluoride.Method The DNA damage and apoptosis were measured after L-02 cell incubated with 80μg/ml sodium fluoride and/or 1.73μg/ml selenium.Results The rates of DNA damage and apoptosis of L-02 cell in the fluoride group were significantly higher than those of the control group,the selenium group and the F-Se group(P<0.01).However,the DNA damage and cell apoptosis rates in the control group,the selenium group,and the F-se group showed no significant difference each other(P>0.05).Conclusion Fluoride exposure induces DNA damage and apoptosis of L-02 cell,and the proper level of selenium can inhibit the DNA damage and cell apoptosis induced by fluoride.
