牡蛎糖蛋白的纯化分离研究

【摘要】 用DEAE-52阴离子交换柱提取、纯化牡蛎糖蛋白,分离纯化的牡蛎糖蛋白命名为糖蛋白GⅠ,糖蛋白GⅡ。用葡聚糖凝胶柱层析分别分析纯化糖蛋白GⅠ,糖蛋白GⅡ,结果表明:牡蛎糖蛋白GⅠ中分离出1种糖蛋白组分GⅠ-1;牡蛎糖蛋白GⅡ纯化出4种糖蛋白组分(GⅡ-1,GⅡ-2,GⅡ-3,GⅡ-4),GⅠ-1为未经洗脱的透过峰,该组分可能是糖蛋白凝聚体,由凝胶电泳分析估算得知GⅠ-1,GⅡ-1,GⅡ-2,GⅡ-3的分子量分别为42.5kDa,100.0kDa,55.3kDa,41.4kDa。

【Abstract】 Oyster glycoprotein was isolated by DEAE-cellulose and the findings showed that there were two peaks appeared as glycoproteins,named as GⅠand GⅡ.Further purification was carried out by SephadexG-200 and only one peak showed for GⅠ,named as GⅠ-1 and appeared before the elute solution was applied.It was suggested that the peak is probably coagulate of glycoproteins.Four peaks appeared for GⅡ,named as GⅡ-1,GⅡ-2,GⅡ-3,GⅡ-4.The molecular weights of glycoproteins were determined by SDS-PAGE,42.5kDa,100.0kDa,55.3kDa,41.4kDa for GⅠ-1,GⅡ-1,GⅡ-2,GⅡ-3 respectively.
