热力法和酶解法提取鱼鳞胶原蛋白的工艺及性质研究

【摘要】 以草鱼鱼鳞为原料,采用热力法和酶解法提取鱼鳞中的胶原蛋白,用正交实验优化提取工艺,分析所得胶原蛋白的性质。酶解法提取草鱼鱼鳞胶原蛋白的最佳工艺条件为:每克蛋白中加入7500U的Alcalase碱性蛋白酶,鱼鳞∶水=1∶20(w/v),60℃水解4h,胶原蛋白的水解度为6.43%。热力法提取的最佳工艺条件为,鱼鳞∶水=1∶20(w/v),121℃提取15min,提取率为45.47%。酶解法提取的胶原蛋白的黏度、吸水性和起泡性大于热力法提取的,但保水性、乳化能力和乳化稳定性及泡沫稳定性小于热力法提取的。

【Abstract】 The fish scale collagen was isolated from grass carp scale by hot water and enzyme hydrolysis extraction respectively.The optimum conditions were determined by orthogonal test,and some functional properties of the collagen obtained from the two optimum conditions were compared.The optimum hydrolysis degree 6.43%,could be obtained from enzyme hydrolysis by hydrolyzing fish scale(fish scale∶water was 1∶20g/mL)with 7500U/g at 60℃ for 4h.The highest yield 45.47%,could be obtained from water extraction by extracting fish scale(fish scale∶water was 1∶20g/mL)at 121℃ for 15min.The collagen isolated by enzyme hydrolysis had higher water absorption,intrinsic viscosity and foam formation,while its water retention,emulsifying property and foam stability were lower than those extracted by hot water.
