大鼠蛛网膜下隙出血后骨髓源性神经干细胞移植对脑组织的修复作用
THE REPAIRING EFFECT OF TRANSPLANTATION OF BONE MARROW-DERIVED NEURAL STEM CELLS ON BRAIN TISSUE OF ADULT RATS AFTER SUBARACHNOID HEMORRHAGE
【摘要】 目的探讨大鼠蛛网膜下隙出血(SAH)后骨髓源性神经干细胞(BMSCs-D-NSCs)移植对脑组织的修复作用。方法成功建立Wistar大鼠SAH模型后,随机分为对照组、DMEM组和BMSCs-D-NSCs组,SAH模型两次采用枕大池注入0.3mL自体血的方法建立,后两组在模型制作后24h于枕大池分别注入30μL的DMEM和BMSCs-D-NSCs。BMSCs-D-NSCs用5-溴脱氧尿嘧啶核苷(BrdU)标记。用神经功能等级评分对移植后大鼠的神经功能进行评估;用免疫组化染色观察BrdU阳性细胞。结果BMSCs-D-NSCs组10~40d神经功能等级评分与其他两组相比,差异有显著统计学意义(F=3.508~4.444,q=3.01~3.67,P<0.05)。在BMSCs-D-NSCs移植后约40d,取脑组织行免疫组织化学染色检查,大脑皮质内可见BrdU阳性细胞存在。结论SAH后移植BMSCs-D-NSCs可以有效地促进大鼠神经功能的恢复;BMSCs-D-NSCs能够分化为神经元从而发挥对SAH后的大脑的修复作用。
【Abstract】 Objective To study the repairing effect of transplantation of bone marrow-derived neural stem cells (BMSCs-D-NSCs) on the brain tissue of rats with subarachnoid hemorrhage (SAH). Methods Forty-five adult Wistar rats were randomly divided into three groups, i.e.control,DMEM (Dulbecco’s Modified Eagle Medium) and BMSCs-D-NSCs. After the models had been made, 30 μL of DMEM or BMSCs-D-NSCs was injected into the cisterna magna of the models of the latter two groups, respectively.The SAH models were induced by injection twice of 0.3 mL autologous blood into the cisterna magna. BMSCs-D-NSCs were labeled with 5-Bromo-2’-deoxyuridine (BrdU).The neurological functional grade score was used to evaluate the nerve function of rats at different intervals following the transplantation.The BrdU positive cells were searched with immunohistochemical staining method. Results Distinguished amelioration in the nerve function was observed in BMSCs-D-NSCs group (F=3.508-4.444,q=3.01-3.67,P<0.05); The BrdU positive cells were detected in the brains of the rats. Conclusion Transplantation of BMSCs-D-NSCs can effectively improve the nerve function of rats with SAH. The new generated neurons differentiated from BMSCs-D-NSCs may exert a repairing influence on the brain after SAH.
