超高效液相色谱(UPLC)用于丹参药材水溶性成分指纹图谱研究

【摘要】 目的:利用超高效液相色谱技术建立丹参药材水溶性成分指纹图谱分析方法。方法:实验采用1.7μm的小粒径填料填充的AcquityUPLCBEHC18色谱柱(2.1×100mm,1.7μm)以0.5%甲酸乙腈(v/v)-0.5%甲酸水(v/v)为流动相梯度洗脱;流速:0.2mL/min;色谱柱柱温:30℃;检测波长:优化选择280nm;进样量:2μL。结果:对12批陕西商洛药材基地产丹参药材进行测定,标定共有峰,并通过国家药典指纹图谱相似度计算软件中"中药色谱指纹图谱相似度评价系统A版"计算出其相似度均在0.95以上。结论:相对于常规HPLC而言,UPLC有更好的分离效率、峰容量以及灵敏度。本方法操作简单,大大缩短了分析时间,重现性、精密度和稳定性良好,可用作丹参药材质量控制。

【Abstract】 This is a study of fingerprints of water-soluble components, in an attempt to establish the fingerprint chromatograms for evaluating the quality of Radix salvia miltiorrhizae using ultra performance chromatograph (UPLC). An analysis is made of the special analytical Acquity UPLC BEH C18 column packed with 1.7μm particals (2.1×100mm,1.7μm), using acetonitrile (containing 0.5% (v/v) formic acid)-water (containing 0.5% (v/v) formic acid) as the mobile phase at a flow rate of 0.2 mL/min. The column temperature is measured 30℃, with a detection wavelength of 280 nm, and an injection volume of 2μL. The reference fingerprints for the extraction of 12 batches of Radix salvia miltiorrhizae are collected from Shangluo cultivation base in Shanxi Province. The chromatograms present a similarity of 0.95, calculated using a professional software named Similarity Evaluation System for Chromatographic Fingerprint of Traditional Chinese Medicine (Chinese Pharmacopoeia Committee Version 2004A). It is concluded that the UPLC system enjoys the merits of a higher speed, sensitivity, and resolution, compared with the conventional HPLC system. The method is simple and reliable, with a greatly shortened analyzing cycle. It can be used to control the quality of Radix salviae miltiorrhizae in an effective manner.
