家蝇幼虫、蛹、成虫抗菌肽的聚丙烯酰胺凝胶电泳分析
Polyacrylamide Gel Electrophoretic Analysis of Antimicrobial Peptides in Musca domestica
【摘要】 目的分析家蝇幼虫、蛹、成虫的抗菌肽成分。方法通过针刺感染大肠杆菌诱导家蝇各虫态产生抗菌肽,再经研磨、离心等步骤提取这些抗菌肽,用改进的聚丙烯酰胺凝胶电泳使其分离。结果家蝇幼虫有7条带,成虫有5条,蛹只有4条。结论家蝇幼虫和成虫的抗菌肽种类较蛹的多,且有一些小分子肽段,可能与其较强的抗菌效果有关;家蝇蛹处于相对静止期,其电泳图中抗菌肽条带较少,无较小的条带,抗菌效果可能较幼虫和成虫的差。
【Abstract】 Objective To analyze the antimicrobial peptides from larvae?pupae and adults of Musca domestica Methods Antimicrobial peptides were induced by infection the insects with bacteria. The peptides were extracted,and analyzed using an improved tricine sodium dodecyl sulfate polyacrylamide gel electrophoresis (Tricine-SDS-PAGE) method. Results A total of 7, 4, and 5 different antimicrobial peptides were identified in the larvae, pupae, and the adults, respectively. Conclusion The presence of a large number of antimicrobial peptides in the larvae and adults is correlated with the resistance of this organism.
