【摘要】 目的观察内皮抑素(endostatin,ES)对小鼠结肠癌移植瘤的治疗作用,并探讨其对血管内皮生长因子(vascular endothelial growth factor,VEGF)表达和微血管密度(MVD)的影响。方法随机将16只结肠癌荷瘤小鼠分为实验组(ES组)与对照组(NS组)。实验组左侧腋部皮下注射ES 6mg/kg,对照组于左侧腋部皮下注射0.9%生理盐水0.2ml,1次/d,连用14d。用药结束后测量肿瘤体积并计算抑瘤率,检测VEGF,计数微血管密度,分析NS组与ES组肿瘤体积、VEGF及微血管密度差异的显著性。结果ES组肿瘤平均体积小于NS组,ES组平均体积为(3892.74±820.14)mm~3,NS组平均体积为(2686.84±1382.24)mm~3,差异有显著性(P<0.05);ES组VECF表达低于NS组(P<0.05);ES组MVD计数低于NS组(P<0.05)。结论内皮抑素能通过直接抑制肿瘤细胞分泌VEGF来减少肿瘤血管的形成达到抑制肿瘤生长的目的。更多还原

【Abstract】 Objective To observe the effect of endostatin(ES)on tumor growth of mice colonic cancer and to explore its effect on the expression of VEGF and MVD.Methods Sixteen BALB/c mouse were randomly divided into two groups:①experimental group:ES(6mg/kg)was injected hypodemically on the left side of armpit,once a day for 14 days;②Control group:0.9% NS 0.2ml was injected hypodemieally on the left side of armpit,once a day for 14 days. The tumors bulk were measured,VEGF,MVD and the tumor inhibition rate(TIR)was detected after injection.The difference of the tumors bulk,VEGF,MVD between the 2 groups were assessed.Results The tumors bulk in the experi- mental group was smaller than that in the control group.The tumors average bulk in the experimental group was(3892. 74±820.14)mm~3,the tumors average bulk in the control group was(2686.84±1382.24)mm~3 ,the difference was sig- nificant(P<0.05).The expression of VEGF and MVD in experimental group were lower than that in control group,and the difference was significant(P<0.05).Conclusions ES can inhibit the angiogenesis of the tumor by inhibiting VEGF directly,so can inhibit the growth of tumor in mice.更多还原
