MALT淋巴瘤中bcl-10与NF-κB的表达及其意义探讨

【摘要】 目的研究在黏膜相关淋巴组织结外边缘区淋巴瘤(extranodal marginal zone B cell lymphoma of mucosa associated lymph-oid tissue,MALT淋巴瘤)中bcl-10核表达和NF-κB/p65活化的关系,探讨bcl-10核表达对MALT淋巴瘤发生的价值。方法收集不同部位MALT淋巴瘤74例(包括29例胃、16例肠、15例眼眶、8例肺、2例唾液腺、1例甲状腺、1例唇、1例胸腺和1例附睾),用免疫组化EnVision法检测bcl-10和NF-κB/p65的表达。结果在74例MALT淋巴瘤中,bcl-10的阳性率为94.6%(70/74),其中胞质阳性率为52.7%(39/74),胞核阳性率为41.9%(31/74);NF-κB/p65的阳性率为100%(74/74),其中细胞质阳性率47.3%(35/74),细胞核和细胞质共同阳性率占52.7%(39/74)。统计结果显示bcl-10核阳性与NF-κB核表达间有显著相关性(P<0.001)。结论在MALT淋巴瘤中,bcl-10核表达对NF-κB活化有显著影响,可能是促进细胞增生、导致肿瘤形成的重要因素。

【Abstract】 Purpose To determine the relationship of nuclear bcl-10 expression and NF-κB/p65 activation in MALT lymphomas, and to discuss its value for tumorigenesis of MALT lymphomas. Methods 74 cases of MALT lymphomas were collected and consisted of 29 gastric, 16 intestinal, 15 ocular adnexal, 8 pulmonary, 2 salivary glands, as well as 4 from other sites. Immunohistochemistry was performed for bcl-10 and NF-κB/p65 antibody by EnVision technique on the formalin-fixed and paraffin-embedded tissues. Results In 74 MALT lymphomas, 70 of 74 cases were bcl-10 positive. Among those positive cases, 39 were stained in the cytoplasm and 31 were in the nucleus. All cases were NF-κB/p65 positive, and 35 were in the cytoplasm, 39 were nucleus with cytoplasmic staining simultaneously. Under the Fisher statistical analysis, bcl-10 nuclear expression was closely related with the NF-κB/p65 activation(P<0.001). Conclusion In MALT lymphomas, bcl-10 nuclear expression may induce NF-κB/p65 activation and makes NF-κB/p65 transport into nucleus from cytoplasm. This should be an important factor for cell hyperplasia and tumorigenesis.
