单核细胞趋化蛋白-1基因-2518G/A多态性与中国湖南地区汉族人群脑梗死关系的研究

【摘要】 目的探讨单核细胞趋化蛋白-1(MCP-1)基因-2518G/A多态性与中国湖南地区汉族人群脑梗死(CI)的关系。方法采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)、DNA测序等方法检测162例CI患者(CI组)和150名健康对照者(NC组)MCP-1基因-2518G/A多态性;采用酶联免疫吸附试验(ELISA)检测两组血清MCP-1水平。结果CI组GG、GA、AA基因型频率分别为39.5%、37.0%、23.5%,G、A等位基因频率分别为58.0%、42.0%;NC组GG、GA、AA基因型频率分别为27.3%、39.3%、33.4%,G、A等位基因频率分别为47.0%、53.0%。CI组GG基因型和G等位基因频率显著高于NC组(均P<0.05)。CI组血清MCP-1含量[(157.68±11.60)pg/ml]明显高于NC组[(131.82±10.72)pg/ml](P<0.05);MCP-1基因-2518G/A多态位点中含G等位基因者(GG+GA)血清MCP-1含量[(149.44±15.71)pg/ml]显著高于非G等位基因携带者(AA)[(134.57±15.84)pg/ml](P<0.05)。结论MCP-1基因-2518G/A位点的G等位基因可能是中国湖南地区汉族人群CI发病的遗传易感基因。携带G等位基因的个体可能通过上调MCP-1表达而增加CI的发病风险。

【Abstract】 Objective To investigate the relationship between monocyte chemoattractant protein-1 (MCP-1) gene -2518G/A polymorphism and cerebral infarction(CI) of Han population in Chinese Hunan district.Methods The -2518G/A polymorphism in MCP-1 gene was detected by polymerase chain reaction-restriction fragment length polymorphism analysis (PCR-RFLP) and DNA sequencing in 162 patients with CI (CI group) and 150 normal controls (NC group). The serum level of MCP-1 in the two groups was determined by enzyme-linked immunosorbent assay (ELISA).Results In CI group, genotypic frequency of GG was 39.5%, GA was 37.0%, and AA was 23.5%. The allele frequency of G was 58.0% and A was 42.0%. In NC group, genotypic frequency of GG was 27.3%, GA was 39.3%, and AA was 33.4%. The allele frequency of G was 47.0% and A was 53.0%. The frequencies of MCP-1 -2518GG genetype and G allele in CI group were significantly higher than those in CI group (all P<0.05). The level of serum MCP-1 in CI group [(157.68±11.60)pg/ml]was significantly higher than that in NC group [(131.82±10.72)pg/ml] (P<0.05). The people with MCP-1 -2518G allele (GG+GA) had higher level of serum MCP-1 [(149.44±15.71)pg/ml]than that only with MCP-1 -2518A allele (AA) [(134.57±15.84)pg/ml](P<0.05).Conclusions The MCP-1 gene -2518G/A G allele may be a genetic risk factor for CI of Han populations in Chinese Hunan Hans.The MCP-1 -2518G allele carrier may has up-regulated the expression of MCP-1 and hence is a higher risk of CI.
