高压氧对新生大鼠缺氧缺血性脑损伤模型NF-κB活化和神经细胞凋亡的影响

【摘要】 目的探讨高压氧(HBO)对新生大鼠缺氧缺血性脑损伤(HIBD)模型神经细胞的凋亡和对NF-κB活化的影响。方法新生7日龄健康SD大鼠随机分为空白对照组(n=16)、假手术组(n=16)、HIBD组(n=17)及HBO组(n=18)。Western-blot检测神经细胞IκBα的表达,了解NF-κB的活化,TUNEL试剂盒检测神经细胞的凋亡。结果脑组织缺氧缺血后可在一定程度上诱导NF-κB的活化,HBO治疗可使NF-κB的活化增强;脑组织缺氧缺血后神经细胞的凋亡明显增加,其阳性凋亡数为368.62±8.25,HBO治疗后神经细胞的阳性凋亡数为278.32±10.34,较HIBD组明显减少(P<0.05)。结论HBO治疗可减轻缺氧缺血性脑损伤后神经细胞的凋亡,机制可能与其诱导了神经细胞NF-κB的活化有关。

【Abstract】 Objectives To study the activation of NF-κB and apoptosis of neural cells in neonatal rats with hypo-xic- ischemic brain damage (HIBD) model treated with 2.0 ATA hyperbaric oxygenation (HBO). Methods SD rats of 7 days old weighted 12~14 g were divided into four groups randomly. They were normal group (n = 16), sham operated group (n = 16), HIBD group (Rice’ s model, n = 17) and HBO group (n = 18). Rats in HBO group were treated with HBO after HIBD model for 1 hour per treatment once a day for 7 days. The expression of IκBα: and apoptosis of neural cell were tested by Western- blot and immunohistochemistry, respectively. Results (1) NF-κB activation can be induced by hypoxic-ischemia to certain degree and such an induction is enhanced by HBO; (2) The apoptosis of neural cells in the brain after hypoxic- ischemia were increased obviously (368.62±8.25), however that were dropped significantly in HBO group (278.32±10.34) (P<0.05) .Conclusions HBO can reduce the neural cell apoptosis in brain with HIBD. The possible neuroprotection mechanism of HBO may relate to the activation of NF-kB.
