木贼提取物对早期AS大鼠血管平滑肌凋亡及Fas、FasL表达的干预

【摘要】 目的研究木贼正丁醇提取物对大鼠动脉粥样硬化早期主动脉平滑肌细胞凋亡及Fas、FasL表达的影响。方法选用雄性SD大鼠,随机分为正常对照、病理模型、阳性药及木贼提取物治疗组。复制食饵性AS早期模型,用木贼提取物预防性给药,以吉非罗齐为阳性药对照。实验9周,流式细胞术定量检测主动脉平滑肌细胞凋亡率及Fas、FasL的表达,光镜下观察主动脉形态学变化。结果木贼正丁醇提取物治疗组平滑肌细胞的凋亡率明显高于模型组(P<0.01),Fas、FasL表达均高于模型组(P<0.01及P<0.05),主动脉损伤程度减轻。结论木贼正丁醇提取物可通过调控AS早期主动脉平滑肌细胞Fas、FasL基因表达,促进平滑肌细胞凋亡,阻断AS进展。

【Abstract】 Objective To study the effect of equisetum n-butanol extract on the apoptosis and Fas/FasL expression of arterial smooth muscle cell(ASMC) in rats with early atherosclerosis(AS).Method Fourty SD rats were divided into four groups: the normal control group,the model group,the gemfibrozil treating group and the equisetum extract treating group.The early AS model was reproduced by high fat diet.The model rats were administered with equisetum extract prophylactically,while gemfibrozil was used as positive control drug.After nine weeks,the rate of apoptosis and the Fas/FasL expression of ASMC were detected by the flow cytometry,and the morphological changes of aorta were observed under the light microscopy.Results The rate of apoptosis and the Fas/FasL expression of ASMCs in the equisetum treating group were both higher than those of the model group(P< 0.01,P<0.05),and the degree of atherosclerotic lesion was less than that of the model group.Conclusion Equisetum extract can regulate the expression of Fas/FasL and promote the apoptosis of ASMC of early AS to prevent the progression of early atherosclerosis.
