壳聚糖协同脐血造血干细胞对小鼠的免疫恢复研究
EXPERIMENTAL STUDY OF EFFECT OF WATER SOLUBLE CHITOSAN AND UCB ON SUB-IETHALLY IRRADIATED IMMUNODEFICIENCY MICE
【摘要】 为了解壳聚糖对脐血移植的影响,在体外培养的基础上,采用MTT法观察水溶性壳聚糖对脐血干细胞的作用,同时在体内检测移植后小鼠造血细胞数量、体液免疫和细胞免疫情况,以了解水溶性壳聚糖对脐血造血干细胞植入的影响。观察结果表明,壳聚糖在体外对脐血造血细胞有扩增作用,与细胞因子相比扩增倍数为11.32;脐血造血细胞在体内移植的同时,壳聚糖有类生长因子作用,可促进免疫抑制小鼠造血重建和免疫恢复。
【Abstract】 To determine the effect of the water-soluble chitosan on the umbilical cord blood transplantation(UCBT) in murine model,on the basis of cell culture in vitro,the effect of different concentration of the water-soluble chitosan on proliferation of stem cells were detected by MTT;and some important indices such as survival rate,hematopoiesis reconstitution,T cell and B cell immunity were recorded and analyzed at the same time.The results show that,CS coordinated with cytokine had better promoting effect on umbilical cord hematopoietic cell in vitro.The proliferation multiple were 3.97 and 11.32,respectively.CS can stimulate the proliferation of UCB,and has synergistic effect when coordinated with the cytokine,promotes the recomstitution of immunity and hematopoiesis rebuild of the immunodeficiency mice.
