水通道蛋白4在大鼠蛛网膜下腔出血后脑水肿组织中的表达及高压氧对其影响的研究
Expression of aquaporin 4 in edematous cerebral tissues of rats after subarachnoid hemorrhage and the effect of hyperbaric oxygen on its expression
【摘要】 目的探讨大鼠蛛网膜下腔出血(SAH)后脑水肿组织中水通道蛋白4(AQP4)的表达变化及高压氧(HBO)治疗对其表达的影响。方法将成年Wistar大鼠84只随机分为对照组(6只)、假手术组(6只)、SAH组(36只)、HBO治疗组(36只),SAH组及HBO治疗组又分为术后1d、2 d、3d、4d、5d、7d共6个亚组,每个亚组各6只。枕大池二次自体注血法制备大鼠SAH模型。Loeffler法进行大鼠神经行为学评分,取出脑组织后用干湿比重法检测脑组织含水量,Western blot方法检测AQP4表达。结果SAH后1d脑含水量和AOP4表达增加,2d即显著增加,2d与3 d差异无显著性,后随时相逐渐递增,7d达到最高。AQP4的表达和脑含水量呈显著正相关(r=0.99,P<0.05)。HBO治疗各亚组AQP4表达水平较相应SAH各亚组均显著降低(P<0.05)。结论AQP4与SAH后脑水肿的形成密切相关,HBO可以下调AOP4的表达,减轻SAH性脑水肿。
【Abstract】 Objective To study the changes in expression of aquaporin 4(AQP4)in edematous cerebral tissues of rats aider subarachnoid hemorrhage(SAH)and the effect of hyperbaric oxygen(HBO)on this expression.Methods A total of 84 adult Wistar rats were randomly allocated into the control group(n=6),sham operation group(n=6),SAH group(n=36)and HBO group(n=36). The SAH and HBO groups were then subdivided into 6 subgroups,respectively at 1,2,3,4,5 and 7d after the treatment,6 rats for each subgroup.SAH models were induced by twice injecting autologous blood into the cisterna magna.Loeffler’s method was applied to record scores of neurological behaviors of rats;the water content of the removed cerebral tissues was determined by the dry-wet weight method; and,the Western blot assay was employed to detect the expression of AQP4.Results The water content of cerebral tissues and AQP4 expression increased at 1d after SAH,significantly increased at 2 d,had no significant differences between 2d and 3d,and then increased gradually in a time-dependent manner until 7d reaching their climaxes.AQP4 expression was positively correlated with the water content of cerebral tissues(r=0.99,P<0.05).The levels of AQP4 expression in HBO subgroups were significantly reduced,compared with that of the corresponding SAH subgroups(P< 0.05).Conclusion AQP4 is closely related with the development of cerebral edema after SAH.HBO may down-regulate the expression of AQP and alleviate cerebral edema after SAH.
