雷公藤内酯醇对佐剂关节炎模型大鼠关节中核因子-κB受体激活剂配基表达的影响
The Effect of Triptolide on Expression of Receptor Activator of Nuclear Factor-κB Ligand and Osteoprotegerin in Joints of Rat Adjuvant Induced Arthritis
【摘要】 目的:探讨雷公藤内酯醇对佐剂关节炎大鼠关节中核因子-κB受体激活剂配基(RANKL)和护骨素(OPG)表达的影响。方法:复制大鼠佐剂关节炎模型,关节炎起病后分别灌胃给予雷公藤内酯醇和甲氨蝶呤,关节炎病程高峰期运用免疫组化和图像分析技术测定关节滑膜和骨组织RANKL和OPG的表达,同时测定胫骨关节端骨密度。结果:给予雷公藤内酯醇和甲氨蝶呤的大鼠骨密度均高于模型组(P<0.05),大鼠关节滑膜、软骨下骨和小梁骨区域中RANKL表达量均低于模型组(P<0.05,P<0.01),OPG关节骨组织表达量均低于模型组(P<0.05)。结论:抑制炎性关节中RANKL表达可能是雷公藤内酯醇和甲氨蝶呤抑制类风湿关节炎关节骨侵蚀的机制之一。
【Abstract】 Objective:To explore the effects of triptolide(TP) on the expression of receptor activator of nuclear factor-κB ligand(RANKL) and osteoprotegerin(OPG) in the joints of rat adjuvant induced arthritis(AA).Methods: AA was induced in Wistar rats.Arthritis rats were orally given TP and methotrexate(MTX) at the onset(day 0) of arthritis.On the peak of arthritis(day 24) the expression of RANKL and OPG in the joints were analyzed using specific immunohistochemistry and image analysis.Bone mineral density(BMD) at the distal end of femoral bone was also evaluated.Results: Compared with AA control(mode 1) group,Arthritis rats treated with TP and MTX had higher BMD(P<0.05)and lower levels expression of RANKL in synovium,subchondral and trabecular bone(P<0.05,P<0.01)respectively.OPG level was lower than that of AA group in subchondral and trabecular bone(P<0.05).Conclusion: TP and MTX may inhibit RA bone erosion by decreasing the expression of RANKL in inflammatory joints.
