【摘要】 目的:观察宣痹汤对Ⅱ型胶原诱导的关节炎(CIA)大鼠Th1/Th2细胞漂移的影响。方法:将大鼠随机分为正常组(N组)和模型复制组,模型复制组经成功复制CIA大鼠模型后,随机分为模型组(M组)和宣痹汤组(X组)。X组灌胃给予宣痹汤3周,同时M组和N组给予9.0 g/L氯化钠注射液3周。其间测量非致炎足踝关节直径,并进行关节炎指数评分。灌胃结束后,经股动脉取全血,使用ELISA法测定血清INFγ和IL-4水平;使用流式细胞术,以外周血CD4+INFγ+和CD4+IL-4+T细胞分别代表Th1和Th2细胞,测定各自在淋巴细胞所占比例,计算Th1/Th2比值,并将CIA大鼠踝关节直径与Th1/Th2比值进行相关性分析。结果:与M组相比,CIA大鼠关节直径、关节炎指数评分均有改善(P<0.05);血清INFγ和IL-4水平,Th1所占比例以及Th1/Th2比值均显著下降(P<0.01),Th2所占比率显著上升(P<0.01),且CIA大鼠临床症状的改善与Th1/Th2比值降低具有相关性(r=0.846,P<0.01)。结论:宣痹汤能有效缓解CIA大鼠临床症状,恢复Th1/Th2漂移,这可能是其临床治疗类风湿关节炎的机制之一。更多还原

【Abstract】 Objective:To investigate the effects of Xuanbi decoction on the Th1/Th2 shift in collagen-induced arthritis(CIA) rats.Methods: The rats were randomly divided into normal(N) and modeling groups.After successfully preparing the CIA rat models,the modeling group was randomly assigned into the model group(M group) and Xuanbi decoction group(X group).Group X was fed intragastrically with Xuanbi decoction for 3 weeks.Both Group M and Group N were treated likewise with normal saline for 3 weeks.During the interval,the diameter of non-sensitization ankle was measured and the arthritis index scored.After the intragastric lavage,the whole blood was collected via the femoral artery and the serum levels of INFγ and IL-4 measured by ELISA;Flow cytometry was employed to count the CD+4 INFγ+ and CD+4 IL-4+ T cells in the peripheral blood.They represented Th1 and Th2 cells respectively.Their percentages in lymphocytes were measured and the ratio of Th1/Th2 calculated.And a correlation analysis between CIA rat ankle diameter and Th1/Th2 ratio was conducted.Results:As compared with those of Group M,both CIA rat ankle diameter and arthritis index improved(P<0.05).The serum levels of INFγ and IL-4,Th1 percentage and Th1/Th2 ratio all decreased markedly(P<0.01).Th2 percentage increased markedly(P<0.01) and the symptomatic improvement of CIA rats were correlated with the decrease of Th1/Th2 ratio(r=0.846,P<0.01).Conclusion:Xuanbi decction could effectively relieve the clinical symptoms of CIA rats and restore the Th1/Th2 shift.This is probably one of underlying mechanisms for clinical treatment of rheumatoid arthritis.更多还原
