survivin mRNA及蛋白在肺癌组织芯片中的表达及意义

【摘要】 目的:探讨survivinmRNA和蛋白的表达在肺癌发生、发展中的作用和意义。方法:利用荧光原位杂交(fluores-cencein situhybridization,FISH)方法、免疫组化SP法和组织芯片(tissue microarray,TMA)技术,检测原发性肺癌和正常肺标本共计240点中survivinmRNA和蛋白的表达情况。结果:原发性肺癌中有66.7%表达survivinmRNA,57.4%表达survivin蛋白,均显著高于正常组(P<0.05);survivinmRNA和蛋白的表达与肺癌分化程度、淋巴结转移和临床分期显著相关(P<0.05)。survivinmRNA和蛋白的表达呈显著正相关(rs=0.583,P<0.05)。结论:survivinmRNA和蛋白在肺癌中高表达,与肺癌进展程度和恶性行为相关,对肺癌的发生、发展起促进作用,可为预后估计和治疗提供一定的依据。

【Abstract】 Objective:To investigate the significance of survivin mRNA and protein expressions in the oncogenesis and progression of lung cancer.Methods:The expressions of survivin mRNA and protein in 240 cores of primary lung cancer and normal lung tissue were detected by fluorescence in situ hybridization(FISH) method,immunohistochemical SP method,and tissue microarray technology.Results:survivin mRNA(66.7%) and protein(57.4%) expressions were significantly higher in primary lung cancer than normal control(P<0.05).The expressions of survivin mRNA and protein were significantly related to the differentiation degree,lymph node metastasis,and clinical stages of lung cancer(P<0.05).The correlation between survivin(mRNA) and protein was significant positive(r_s=0.583,P<0.05).Conclusion: survivin mRNA and protein were highly expressed in lung cancer.It was related to the progress and malignant behavior of lung cancer.survivin accelerated oncogenesis and progression of lung cancer which provided basis for clinical therapy and prognosis.
