应用荧光定量聚合酶链反应产前诊断巨细胞病毒感染

【摘要】 目的探讨FQ-PCR产前诊断巨细胞病毒感染的临床应用价值。方法用FQ-PCR对20例巨细胞病毒感染孕妇的胎儿进行产前诊断,同时对孕妇感染CMV-DNA拷贝数的高低与发生垂直传播的关系进行了探讨。结果4例胎儿绒毛组织CMV-DNA阳性,阳性率为20%(4/20),CMV-DNA平均为5.0×106,±2.2×106拷贝/ml:16羊水标本阴性,CMV-DNA<25拷贝/ml,新生儿血、尿液CMV-DNA均阴性,产后临床追踪观察3年,无CMV感染迹象。孕妇感染CMV-DNA拷贝数的几何均数为8.0251±0.3986时,有可能发生垂直传播(t=16.24,P<0.01)。结论FQ-PCR产前诊断巨细胞病毒感染结果可靠,有一定的临床应用价值。

【Abstract】 Objective:To evuluate FQ-PCR on detection cytomegalovirus in prenatal diagnosis. Methods:Fetal diagnosis was made by fluorescence quentitative PCR. For amplification of CMV-DNA with CVS(n=4) and amniotic fluid(n=16). Results: The positive rate of CMV-DNA was 20%(4/20)by FQ-PCR. The average copies of CMV-DNA was 5.0×106±2.2×106 copies/ml. The CMV-DNA of aminiotic fluid, blood and urine of infant was negative. The copies of negative cases were<25 copies/ml. The geometric mean of CMV-DNA in prenagnat women with cytomegalovirus infection was 8.0251±0.3986 that could be associated with vertical transmission. Conclusion: FQ-PCR is a valuabl method for detection cytomegalovirus in prenatal diagnosis.
