通心络对病毒性心肌炎慢性期心肌纤维化作用的实验研究

【摘要】 目的:观察通心络对病毒性心肌炎慢性期心肌纤维化小鼠的抗心肌纤维化作用,为扩大通心络的应用范围提供重要的实验依据。方法:建立病毒性心肌炎慢性期心肌纤维化小鼠模型,通过透射电镜观察心肌细胞超微结构;HE染色、苦味酸天狼星红染色观察心肌胶原数量;放免法测定血清透明质酸(HA)的含量。结果:电镜、HE染色及苦味酸天狼星红染色示通心络各剂量组胶原较模型组均有不同程度的减少,胶原容积积分示模型组和高剂量组形成显著性差异(P<0.05)。血清HA模型对照组较正常对照组明显升高,有显著性差异(P<0.01),但各剂量组与模型组比较均没有显著性差异。结论:通心络各剂量组均有抗心肌纤维化作用,其中以高剂量组效果最为明显。

【Abstract】 Objective:To observe the anti-fibrosis effect of Tong xin luo on myocardial fibrosis in chronic stage of viral myocarditis,and offer important experiment gist to enlarge application of Tong xin luo.Methods:Establish the model of myocardial fibrosis in chronic stage of viral myocarditis,observe the ultrastructure of myocardial cell with tramission electron microscope,the quantity of myocardial collagen was observed by staining with HE and Sirus red in supersaturated picric acid solution;the amount of HA in serum was measured with RIA.Results:Myocardial collagen in model group increased obviously compared with normal group from the results of electron microscope,HE,Sirus red in supersaturated picric acid solution.Myocardial collagen in every dose group decreased to different degree and there was significant difference in PCV of model group compared with high dose group(P﹤0.05).There was significant difference in the amount of HA in serum of model group compared with normal group(P﹤0.01),but no significant difference compared with every dose group.Conclusion:Tong xin luo every dose group could resist myocardial fibrosis,but effect of high dose group was most obvious.
