水稻纹枯病发病过程PR1和PBZ1的表达动态

【摘要】 采用从福建省稻田分离纯化的纹枯病菌(Rhizoctonia so lani)菌株FJ-15接种籼稻9311,分析了水稻在纹枯病菌侵染致病过程中,编码病程相关蛋白的基因表达动态,并观察了症状的变化。Northern b lot分析表明:PR1在接种12 h后开始表达,在之后的4个时间段其表达量逐步增强;而PBZ1也在12 h开始表达,在48 h表达量激剧增强几乎与72 h表达量相当。组织和症状观察表明,接种12 h后叶鞘表面菌丝纵横分枝,接种36 h后出现零星病斑,接种48 h后表现典型的受害症状,接种72 h后病斑继续扩大,并可蔓延到非接种叶鞘。结果表明,PR1和PBZ1的表达与水稻和纹枯病菌亲和互作的过程存在对应关系。

【Abstract】 To investigate the pathogenesis-related gene(PR) expression through the infection process of rice sheath blight,rice cultivar 9311 was inoculated with the fungal pathogen Rhizoctonia solani isolated and purified from the breeding field of Fujian Agriculture and Forestry University in Fuzhou,China.Rice samples were collected at 0,4,12,24,36,48,72 h post-inoculation for total RNA extraction and observation of di-(sease) development.Two PR gene fragments,PR1 and PBZ1,were used as probes for Northern blot analysis.The results showed that expression of both PR1 and PBZ1 was induced at 12 h post-inoculation,however PR1 was increased gradually from 12 h to 72 h post-inoculation,while PBZ1 drastically increased at 48 h post-ino-(culation) to almost the maximum of 72 h post-inoculation.Analysis of infection process showed that mycelium grew randomly on the surface of tissue at 12 h post-inoculation,few lesions started to come out at 36 h post-(inoculation),and typical lesions were found in all plants at 48 h post-inoculation.Integrating the results of mole-(cular) analysis and infection progress observation,we concluded that expression of PR1 and PBZ1 may be rela-(ted) with disease development of rice sheath blight,which may suggest that SA signal is involved in the defense pathway.
