小鼠肾小体发育过程中的细胞凋亡及Bcl-2和Bax基因表达
Apoptosis and Expression of Bcl-2 and Bax During Developing Renal Corpuscle in Mouse
【摘要】 目的探讨细胞凋亡及凋亡相关基因bcl-2和bax在小鼠不同发育阶段肾小体中表达的变化特征及其可能的生物学意义。方法采用末端脱氧核苷酸转移酶介导的缺口末端标记法(TUNEL染色法)及bcl-2和bax蛋白免疫组织化学方法,对发育不同阶段小鼠肾小体的细胞凋亡及Bcl-2和Bax基因蛋白表达进行观察。结果在小鼠肾小体发育的不同阶段均有TUNEL阳性细胞的表达,凋亡阳性细胞多出现在Ⅲ、Ⅳ期肾小体内,而此时bcl-2/bax比值明显降低。结论细胞凋亡高峰主要出现在Ⅲ、Ⅳ期肾小体,bcl-2/bax比值的降低可能是这一现象发生的重要机制之一。
【Abstract】 Objective To observe apoptosis and the expression of bcl-2 and bax gene during the developing renal corpuscle in mouse and understand their potentially biological significance.Method Terminal deoxynucleotidyl transferase mediated dUTP-biotin nick endlabeling(TUNEL staining method),bcl-2 and bax immunohistochemisty method were used.Rerult The expression of TUNEL positive cells could be easily observed and identified in Ⅲ and Ⅳ stages;but the radio of bcl-2 and bax was decreased.Conclusion apoptosis are necessary for development of renal corpuscle.One important mechanism is that the decrease of bcl-2/bax ratio causes this phenomena.
