水流动力学注射方法对小鼠肝脏的毒性

【摘要】 目的评价水流动力学注射方法对小鼠肝脏的毒性。方法通过小鼠尾静脉快速注射1.7 ml生理盐水溶液,于注射后不同时间点取血检测ALT及AST水平并取肝脏进行形态学观察。结果注射10 h后,ALT及AST水平最高,24 h后迅速下降,7天后恢复到正常水平。8 s注射组注射后10 h光镜下未见明显病理改变,电镜下可见线粒体轻微肿胀,3天后肝细胞超微结构恢复正常。3 s注射组注射后10 h表现为肝窦明显扩张充血,电镜下可见大部分线粒体内出现絮状沉积物,24 h出现小片坏死区域,7天后基本恢复到正常结构。结论水流动力学注射对肝脏的损伤程度与注射速度有关,损伤可在1周内迅速恢复。

【Abstract】 Objective To evaluate the toxicity of hydrodynamics-based procedure to the liver of mice.Methods 1.7 ml of saline was rapidly injected into mice by tail vein.The blood was taken at different time points for determination of ALT and AST.The livers were taken for morphologic evaluation.Results The highest value of ALT and AST was observed at 10 h and declined sharply at 24 h,fell to the normal values at day 7 after injection.No obvious pathological changes were seen with light microscope and slight mitochondrial swelling with electron microscope were found in mice with 8 seconds of injection at 10 h after injection,the normal ultra-structure of liver was restored at day 3 after injection.In mice with 3 seconds of injection,the congestive hepatic sinus at 10 h and small necrotic foci at 24 h with light microscope were respectively observed after injection.Under electron microscope,woolly focal densities in most mitochondrias were found in hepatocytes.The normal histological structure was restored at day 7 after injection.Conclusion The injury of hydrodynamics-based procedure to liver is associated with injection velocity and it could be rapidly restored within one week.
