【摘要】 目的:制备糖尿病胃轻瘫大鼠模型,通过检测其胃壁组织SCF的表达水平,探讨SCF与糖尿病胃轻瘫的关系。方法:用链脲佐菌素(Streptozocin,STZ)建立糖尿病胃轻瘫大鼠模型,应用RT-PCR和Wester blotting方法分别检测各实验组胃壁SCF基因(cDNA)及蛋白质的表达水平,分析SCF表达水平与糖尿病胃轻瘫的关系。结果:糖尿病胃轻瘫大鼠胃排空明显延迟;SCF基因和蛋白的表达水平明显降低;用西沙必利处理后表达升高,且与病情轻重有相关性。结论:SCF与糖尿病胃轻瘫发病有关,SCF在糖尿病胃轻瘫中的表达水平可以反映其病情发展过程。更多还原

【Abstract】 Objective: To model diabetic gastroparesis(DGP) in rats and evaluate the role of SCF in the diabetic gastroparesis rats by detecting the level of stem cell factor(SCF) in gastric tissue of diabetic gastroparesis rats in order to discuss the relationship between SCF and DGP.Methods: To model diabetic gastroparesis in rats with intraperitoneal injection of streptozocin(STZ).The models were treated with cisapride for two months,and then the function of gastric emptying of all rats and SCF levels,including mRNA and protein in gastric walls,were detected through reverse transcription polymerase chain reaction(RT-PCR) and western-blotting(WB).Results: The function of gastric emptying was delayed and SCF levels of DGP rats were decreased obviousely.After treated with cisapride,SCF levels,however,were increased,dramatically under the levels of control rats.Conclusion: The stage of DGP seems closely associated with SCF levels in gastric walls of animal models.There may be positive correlation between SCF levels and function of gastric emptying.更多还原
