青蒿琥酯抑制MCF-7细胞的机制探讨
Effects of Artesunate and Its Mechanism on MCF-7 Cells
【摘要】 目的探讨青蒿琥酯(Artesunate,ART)对乳腺癌细胞MCF-7抑制作用及其机制。方法ART处理细胞3天后,采用MTT比色法检测细胞增殖功能,观察细胞形态、结构的改变,免疫细胞化学检测bax、nm23、PCNA、VEGF、bcl-2蛋白的表达情况。结果ART对乳腺癌细胞MCF-7的抑制作用呈明显浓度依赖性,可导致细胞形态、结构的改变;免疫细胞化学结果显示20μmol/L青蒿琥酯作用两种细胞72小时后,可上调bax、nm23,下调PCNA、VEGF蛋白的表达,bcl-2蛋白表达无明显变化。结论ART有抑制MCF-7细胞增殖的作用,其机制可能与上调bax、nm23,下调PCNA、VEGF蛋白的表达有关。
【Abstract】 Objective To study the effects of artesunate(ART)and its mechanism on MCF-7 cells.Methods After treated MCF-7 cells with ART for 3 days,the proliferation of MCF-7 cells were examined with MTT-assay,meanwhile the morphological change and the expression of PCNA,bcl-2,bax and nm23 was observed.Results MCF-7 cells were treated with ART for 3d,led to a obviously dose dependent inhibition.ART could change the morphology and ultra structure of MCF-7 cells significantly.Immunocytochemical staining indicated treatment with artesunate(20 umol/L) for 72 hours could be upregulate the expression of bax、nm23,downregulate the expression of PCNA and VEGF protein; not change the expression of bcl-2 in MCF-7 cells.Conclusion Artesunate showed obvious anticancer activity on MCF-7 cells.The mechanism on anticancer effect of artesunate may be related to up-regulation the expression of bax、nm23,down-regulation the expression of PCNA and VEGF protein.
