人脑外伤后皮层核因子-κB的表达
Cortical expression of nuclear factor-kB after human traumatic brain injury
【摘要】 目的探讨核因子-κB(nuclear factor-κB,NF-κB)在人创伤性脑损伤(traumatic brain injury,TBI)后挫伤皮层中的表达情况,包括表达位置、表达强度和表达时相。方法挫伤区皮层的标本来自24例TBI患者,取样时间为伤后5 h~5 d,利用凝胶电泳迁移分析法(electro- phoretic mobility shift assay,EMSA)和免疫组化技术测定NF-κB的活性和表达强度。结果在人TBI后的挫伤区皮层中,NF-κB表达明显上调,表达高峰为伤后48~72 h,NF-κB P65主要在血管内皮细胞和神经胶质细胞中表达,NF-κB P50主要在神经胶质细胞和少量神经元中表达,NF -κB P65的表达强度高于NF-κB P50。结论NF-κB在人TBI后的挫伤区皮层中表达上调,提示其可能在TBI后的病理生理过程中起着重要作用。
【Abstract】 Objective To analyze the expression of nuclear factor kappa B (NF-kB) in contused human brain tissue to identify the expressive location,quantitation and time points.Methods Con- tused brain tissue biopsies were obtained in 24 patients who underwent surgery treatment for brain contu- sions 5 hours to 5 days after trauma.The expression and binding activity of NF-κB were detected by elec- trophoretic mobility shift assay (EMSA) and immunohistochemistry respectively.Results The result showed an immediate,strong and persistent up-regulation of NF-κB in the area surrounding the injured brain as early as 5 hours after injury and it kept at a high level up to 5 days from injury.The maximal ex- pression of NF-κB was detected at 48 to 72 hours after injury,The expression of NF-κB P65 which was mainly located at vascular endothelial cells and neuroglial cells was stronger than that of NF-κB P50 which was located at neuroglial cells and a few neurons.Conclusion NF-κB is up-regulated obviously in the early phase of human brain contusion.NF-κB might play important roles in the pathophysiological course after traumatic brain injury.
