地榆促造血作用的有效部位筛选

【摘要】 目的:筛选地榆促造血作用的有效部位。方法:小鼠骨髓造血细胞培养在96孔培养板中,加入不同浓度的地榆提取物培养10 d,用ATP/生物发光法检测骨髓细胞增殖,体外筛选地榆促造血作用的有效部位;用1、0.5、0.1 mg.kg-1,地榆有效部位连续灌胃给予小鼠14 d,分别计数骨髓有核细胞、外周血白细胞、红细胞和血小板,进一步验证体外筛选所获有效部位的促造血作用。结果:地榆皂苷显示增强细胞因子刺激的小鼠骨髓细胞体外增殖,鞣质和黄酮类成分没有作用;以1、0.5、0.1 mg.kg-1总皂苷处理小鼠14 d后,显著增加了小鼠骨髓有核细胞数量,外周血白细胞、红细胞以及血小板数量也显著升高。结论:地榆皂苷是促小鼠骨髓细胞体外增殖的主要活性部位,地榆皂苷也能升高骨髓抑制小鼠骨髓有核细胞和外周血白细胞、红细胞以及血小板的数量。

【Abstract】 AIM: To screen the active fraction from Sanguisorba officinalis L.for their hematopoietic effect.METHOD:The hematopoietic cells isolated from murine bone marrow(BM) were cultured in vitro,and then were treated with three extracts.The proliferation of BM cells were measured by ATP bioluminescence assay.The hematopoietic effect of the active fraction found in vitro was reconfirmed in hematopoietic inhibition mice induced by cyclophosphamide.Oral administration was performed with gavage in 1、 0.5、(0.1 mg·kg-1) doses of the fraction for 14 days.The BM mononuclear cells and peripheral blood cells including WBC,RBC,and PLT were counted,respectively.RESULTS:In vitro,the fraction containing saponins showed a significant increase in the proliferation of BM cells stimulated by cytokines,whereas tannins and flevones has no effect.In the animal experiment,oral administration of the saponins to cyclophosphamide-induced hematopoietic inhibition mice for 14 days results in increase of the number BM cells,leucocytes,erythrocytes,and platelets.CONCLUSION:The saponins are the active fraction in promoting the proliferation of hematopoietic cell in vitro.The saponins also increase the number of the BM and peripheral blood cells of the hematopoietic inhibition mice induced by cyclophosphamide,in which the increase of RBC and PLT counts is reported for the first time.
