SDS对组蛋白与DNA共振散射光谱影响
Effect of SDS on resonance light-scattering spectrum of histone and DNA complex
【摘要】 目的研究十二烷基磺酸钠(SDS)对组蛋白与DNA共振光散射强度的影响,探讨三者相互作用规律。方法pH 7.5的Tris-HCl缓冲液体系中,将组蛋白与DNA及SDS混合。在入射光波长和荧光波长相同条件下,用荧光分光光度计测定混合液的共振光散射强度。结果扫描光谱显示最大光散射波长为302nm,最佳pH值为7.5,SDS浓度在0.02~0.15mmol/L范围内,体系共振光散射强度达到最大值且趋于稳定。结论小剂量SDS能增强组蛋白-DNA共振光散射体系的光散射强度,反之,大剂量SDS促使组蛋白与DNA分离,使光散射强度大大减弱。
【Abstract】 Objective To study the effect exerted by sodium dodecyl sulfate(SDS) on the resonance light-scattering based on histone and DNA complex and to explore the orderliness of interaction in the three compounds.Methods Histone,SDS and DNA were mixed into the Tris-HCl buffer solution(pH 7.5).Common spectrofluoromenter was used to measure the resonance light-scattering intensity of the mixture with same excitation and emission wavelength.Results Scan spectrum showed that the maximal light-scattering wavelength was 302nm,and the optimum measurement for light-scattering intensity was at pH 7.5.The light-scattering signal was strong in the range of 0.02~0.15?mmol/L SDS.Conclusion Low dose of SDS improved the resonance lightscattering intensity of histone and DNA.Otherwise,high dose of SDS urged the segregation of histone and DNA,so cut down the resonance light-scattering intensity greatly.
