三七总皂苷中人参皂苷Rg1体外透皮吸收的实验研究

【摘要】 目的:研究三七总皂苷(PNS)中主要成分人参皂苷Rg1的透皮吸收特性。考察不同浓度三七总皂苷、不同pH值及一些常用渗透促进剂对其透皮特性的影响。方法:采用V-C扩散池进行体外扩散实验,选择离体大鼠背部皮肤为渗透屏障,HPLC检测法测定Rg1的含量。结果:三七总皂苷的主要成分Rg1可透过大鼠背部皮肤,但渗透系数较小;在一定浓度范围内,其渗透系数随其浓度增加而减小;10%乙醇、10%丙二醇、20%丙二醇、30%丙二醇、2%Azone、2%桉叶油及5%油酸均对其有显著促渗透作用。结论:本研究将为含三七总皂苷经皮给药制剂研究制备提供实验依据。

【Abstract】 AIM: To study the percutaneous absorption of Rg1 in Panax notoginseng and the effects of some commom enhancer and pH value on it. METHODS: Drug permeation tests were performed through excised rat skin in V-C diffusion cell.Rg1 concentration in samples was determined by HPLC. RESULTS: Rg1 could penetrate through excised rat skin with lower permeability coefficient;in the range of concentration 300 mg/mL~60 mg/mL permeability coefficient of Rg1 decreased as its concentration increasd;Among enhancers,10% ethanol,10%,20% and 30% propylene glycol,2% Azone,2% Eucalyptus Oil and 5% Oleic Acid had markedly promotive effect on transdermal penetration of Rg1. CONCLUSION: The research will provide experimental data for skin administration of Chinese medicine containing Panax notoginseng.
