高效液相色谱法测定紫杉醇聚乳酸纳米粒的含量

【摘要】 目的:建立紫杉醇聚乳酸钠米粒含量测定的方法。方法:采用反相高效液相色谱法,以ODS C18柱为固定相,甲醇-水(75:25)为流动相,检测波长为227 nm,流速为1.0 mL/min。结果:回归方程为A=0.648 798 C-0.027 076,r=0.999 9;总的紫杉醇浓度及游离紫杉醇浓度的测定方法的平均回收率分别为(100.42±3.99)%(n=9)和(100.51±2.01)%(n=9);日内精密度RSD≤1.75%,日间精密度 RSD≤1.23%;纳米粒胶体中药物含量为(34.83±2.64)μg/mL,包封率为(99.30±0.10)%。结论:该方法可用于紫杉醇聚乳酸纳米粒的含量测定。

【Abstract】 Objective: To determine paclitaxel(TAX) in paclitaxei - loaded PLA nanoparticles. Methods: An RP-HPLC was developed, the paclitaxel was separated on octadecylsilane column using a mobile phase of methanol water(75 : 25) with detection at 227 nm, the flow rate was 1.0 mL/min. Results: The standard curve was A =0. 648 798C-0. 027 076, r=0. 9999, the average recovery rate of the total drug and the free drug in the nanoparticle colloid solution were (100. 42±3.99)% ( n =9) and (100. 51±2. 01)% ( n =9) respectively. And the precision within day was less than or equal to 1.75%, the precision between day was less than or equal to 1.23%. And the paclitaxel concentration in the nanoparticle colloid solution was (34. 83±2. 64)μg/mL, the extent of entrapment of the nanoparticles was (99. 30±0.10)%. Conclusion: The method could be used to quantitate paclotaxel.
