【摘要】 目的构建超大容量非免疫人源性Fab抗体库。方法通过RT-PCR从208名不同人的外周血淋巴细胞中扩增出人IgG重链Fd段和κ、λ两轻链基因片段,将其克隆入噬粒pComb3中,构建人源性Fab段噬菌体抗体库。结果成功地构建了库容量为2.6×1012的人源性Fab段噬菌体抗体库。结论新的噬菌体抗体库对筛选不同抗原位点的抗体,提供有用的资源。更多还原

【Abstract】 Objective To construct a human Fab phage antibody library.Methods Fd heavy chain gene and κ、λ light chains gene of IgG obtained by RT-PCR from 208 samples of different human lymphocytes were cloned into p Comb3 vector and the human Fab phage anti-body library was constructed.Results A human Fab phage antibody library with 2.6×10¹² capability was successfully constructed.Conclusion This new phage antibody library is set to become a valuable source of antibodies to many different targets.更多还原
