玉米DUS测试标准品种的SSR分子指纹图谱的构建

【摘要】 研究了39个玉米DUS测试标准品种的SSR分子指纹图谱的构建方法。结果显示:①用改良CTAB法得到的玉米DNA的A260/A280为1.82,A260/A230为2.02,其条带清晰,降解少,质量明显优于SDS法;②从所用的国内外63对核心引物中筛选出PIC值高、条带清晰、稳定性好的20对玉米SSR核心引物;③用8%聚丙烯酰胺电泳在20对引物扩增产物中检测出了171个位点,PIC值为0.855 4,远远高于3%琼脂糖电泳,更适合DUS测试;④建立了DUS标准品种SSR分子指纹图谱,为DNA检测方法在玉米新品种DUS测试中的运用奠定了基础。

【Abstract】 This paper studied the building method on DNA digital fingerprints map of 39 maize DUS testing standard inbred lines.The results showed that purity(A260/A280=1.82;A260/A230=2.02) and quality of maize DNA by improvement CTAB method was better than it by SDS method;20 pairs core primers out of 63 pairs of core SSR primers which had been used by other labs were selected which gave stable and high polymorphism information content(PIC);8% polyacrylamide gel was fitter for DUS testing than 3% Metaphor agarose gel.Among the same amplification yield,171 alleles and higher PIC value(0.855 4) were detected by 8% polyacrylamide gel which was over higher than it by 3% Metaphor agarose gel;Based on amplification bands with the 20 pairs core primers,built DNA digital fingerprinting mapping of DUS testing standard inbred lines,built base for molecular screening of DUS testing of new maize cultivars.
