姜黄素对晶状体上皮细胞增殖的抑制作用
Inhibitory effect of curcumin on proliferation of lens epithelial cells
【摘要】 目的探讨姜黄素(Cur)对表皮生长因子(EGF)诱导的牛晶状体上皮细胞(LEC)增殖的抑制作用及其量效与时效关系。方法MTT测定不同浓度的Cur作用于牛LEC不同时间后细胞增殖的情况,流式细胞术(FCM)检测不同浓度的Cur作用于牛LEC不同时间后增殖细胞核抗原(PCNA)的表达状况。结果(1)MTT法结果表明,高、中、低不同浓度的Cur作用24h后,其增殖抑制率分别为66.65%、22.43%、8.72%,呈明显的剂量-效应关系,20mg/L的Cur作用6、12、24、48、72h后,其增殖抑制率分别为9.97%、19.81%、22.43%、41.67%、81.59%,呈明显的时间-效应关系。(2)FCM检测表明,Cur对LEC内PCNA蛋白表达有明显的下调作用,也呈明显的时间-效应关系和剂量-效应关系。结论Cur能有效抑制EGF诱导的LEC增殖,可望成为防治后发性白内障的理想药物。
【Abstract】 ObjectiveCurcumin(Cur) play an important role in the inhibition of the proliferation of tumor cells.This paper was to investigate the inhibitory effects of curcumin on proliferation of cultured bovine lens epithelial cells (LECs) induced by recombinant human epidermal growth factor (rhEGF).MethodsPrimary cultured bovine LECs were used in the study.Proliferation of LEC was induced by 50μg/L rhEGF,in the same time,30,20,10mg/L Cur were added to the LECs culture separately for 24 hours.The inhibition of proliferation of bovine LEC was detected by methyl thiazolyl tetrazolium (MTT).The expression of proliferating cell nuclear antigen (PCNA) in LECs was analyzed by flow cytometer (FCM).ResultsAfter addition of 30mg/L,20mg/L and 10mg/L of Cur for 24 hours,proliferation of LECs were inhibited by 66.65% at concentration of 30mg/L, and by 22.4 3% at 20mg/L and 8.72% at 10mg/L of Cur.The positive ratio of PCNA staining was (33.17±1.20)%,(40.86±2.16)% and (58.88±1.56)%,respectively.LECs were treated with 20mg/L Cur for 6,12,24,48 and 72 hours,cells proliferation was inhibited by 9.97%,19.81%,22.43%,41.67%,81.59% and the positive ratio of PCNA staining was(53.31±2.35)%,(55.12±2.13)%,(40.86±2.16)%,(28.13±4.15)%,(12.58±3.61)%,respectively.ConclusionCur can decrease proliferation of LECs induced by rhEGF at a dose-dependent and time-dependent manner.PCNA expression in LECs is down-regulated by Cur.The Cur may become a potential drug to prevent and cure after cataract.
