PESV对荷Lewis肺癌小鼠血流变学的影响的实验研究

【摘要】 目的观察蝎毒多肽提取物(PESV)抑制荷Lewis肺癌C57/B16小鼠肺癌生长及对荷瘤小鼠血流变学的影响。方法将0.2mL Lewis肺癌细胞悬液给C57/B16小鼠尾静脉注射,8d后将小鼠随机分组,治疗组用PESV腹腔注射,对照组用0. 9%生理盐水腹腔注射,隔日1次,共10次。第30d取全血做血流变学分析,并处死小鼠,取小鼠肺脏,进行分析。结果 PESV抑制荷Lewis肺癌小鼠肺转移作用明显(治疗组和对照组肺肿瘤数为3.2±1.2和13.5±5.3,P<0.01),血流变学分析显示,与对照组相比,PESV组血流变学指标均有不同程度的降低,尤其是全血低切表观粘度、全血高切表观粘度、 Casson粘度、Casson屈服应力,其指标明显低于对照组(P<0.01)。结论 PESV对荷Lewis肺癌的C57/B16小鼠肺转移具有抑制作用,可能通过降低血液粘度实现对Lewis肺癌的抑制作用。

【Abstract】 Objective To confirm antimetastatic the effect of PESV on Lewis lung carcinoma in C57/B16 mice and the change of PESV on the hemorheology in the whole blood. Methods 0.2mL Lewis cell suspension was injected into the tail of C57/B16 mice.8 days later, mice were divided into two groups randomly, treatment group was administered by intraperitoneal injection of PESV, and control group by intraperitoneal injection by 0.9% NS solmion,once every other day, after 30 days,blood to drawed test hemorheological values and analysize the lung of dead mice. Results There was significant antimetasitic effect on Lewis lung carcinoma in C57/B16 mice(the number of lung metastatic foci in the treatment group and the control group was 3.2 ± 1.2 and 13.5 ± 5.3,P<0.01 );After the use of PESV, there are significant differences between PESV group and control group, especially the whole blood viscosity at 3s-1、 200s-1 shear rate(P<0.01 vs control). Conclusion PESV can inhibit metastasis of Lewis lung carcinoma in C57/B16 mice probably through hemorheological changes.
