氟伐他汀对阿霉素肾病大鼠SOD和MDA水平的影响
THE EFFECT OF FLUVASTATIN ON THE SOD AND MDA IN ADRIBLASTINE NEPHROSIS RATS
【摘要】 目的:探讨氟伐他汀对阿霉素肾病大鼠肾皮质匀浆丙二醛(MDA),过氧化物歧化酶(SOD)水平、血浆蛋白和血脂水平的影响及其与临床的关系。方法:单次尾静脉注射阿霉素法制备阿霉素肾病大鼠模型,硫代巴比妥酸(TBA)法测定肾皮质匀浆液丙三醛(MDA)含量,邻苯三酚自氧化法测定肾皮质匀浆超氧化物岐化酶(SOD)。结果:①注射阿霉素2周后,电镜示肾小球基膜增厚,模糊不清,足突融合,内皮细胞呈连拱状。②氟伐他汀治疗后,血总蛋白、白蛋白、甘油三酯、低密度脂蛋白等与阿霉素肾病组相比较均差异显著。③氟伐他汀治疗组大鼠肾匀浆SOD水平(3.32±0.26)mmol/L高于阿霉素组(2.14±0.74)mmol/L;MDA水平(2.02±0.17)mmol/L低于阿霉素组(2.85±0.37)mmol/L,2者相比差异显著(P<0.01)。结论:阿霉素可致大鼠脂质代谢异常和脂质过氧化,而在电镜下可见氟伐他汀可以降低其血脂水平并在一定程度上有抗氧化作用。
【Abstract】 Objective:To investigate the effect of Fluvastatin on renal cortex homogenate MDA,SOD,plasma protein and blood lipids and to explore their relationship with the clinical treatment.Methods:Model of rats were established by intravenously injection of VAM in caudal vein,Renal cortex homogenate MDA content were measured by TBA method and SOD was detected by auto-oxidation of pyrogallic acid.Results:①Two weeks after injection adriblastine,the glomerular basement membrane increased thickness and unclearly,foot process became confluence,endothelial cell were jointed as arch.②There was significantly difference in TP,ALB,TG,TC etc.between the treatment group with Fluvastatin and adriblastine group.③The level of renal cortex homogenate MDA in Fluvastatin group(3.32h0.26 mmol/L)was higher than that in ADM group(2.14±0.74 mmol/L),while the level of MDA(2.02±0.17 mmol/L)was lower than that of ADM group(2.85±0.37 mmol/L).The difference between them was significant(P<0.01).Conclusion:Adriblastine can lead to lipid metabolism disturbance and lipid dioxide,Fluvastatin may decrease the blood lipids and have some extent anti-oxidation effect,which can be confirmed by the electron microscope.
