TK基因突变试验检测叶绿素的抗诱变性
DETECTION OF ANTIMUTAGENICITY OF CHLOROPHYLL BY TK GENE MUTATION TEST
【摘要】 目的:用人的类淋巴母细胞TK6的TK基因突变试验检测叶绿素的抗诱变性。方法:不加S9活化的条件下,用叶绿素10、20、40、80μg/ml与阳性致突变物丝裂霉素C(MMC)0.5μg/ml同时处理TK6细胞3 h,采用微孔板法进行平板接种效率和突变频率的测定。结果:随受试物剂量的增加,叶绿素对MMC诱变性的拮抗作用增大,当剂量达到40μg/ml以上时总突变频率较MMC组有显著降低(P<0.05)。结论:叶绿素具有拮抗MMC诱导的tk基因突变的作用。
【Abstract】 Objective:To detect and evaluate the antimutagenicity of chlorophyll by TK gene mutation test.Methods:The TK6 human lymphoid cells were exposed to chlorophyll at concentrations 10,20,40,80 μg/ml simultaneously,with 0.5 μg/ml MMC for 3 hours without S9.Determination of plating efficiency and mutant frequency were performed by using the microwell method.Results:A dose-dependent inhibiting effect of chlorophyll on MMC mutagenicity was found,and the mutant frequency depressed significantly compared with the MMC control when the concentration of chlorophyll arrived at 40 μg/ml(P<0.05).Conclusion:chlorophyll has obvious inhibiting effect on TK gene mutation induced by MMC.
