低氧状态下缺氧诱导因子信号转导途径对糖酵解的作用

【摘要】 目的:总结并分析缺氧诱导因子代谢途径及对糖酵解酶的调节,探讨长时间大强度的运动或者高原训练的可能机制,以消除大强度运动对机体的伤害。资料来源:应用计算机检索Medline1993-01/2004-12关于缺氧诱导因子的文章。检索词“Hypoxia-induciblefactor1”。同时利用计算机检索中国期刊全文数据库1993-01/2004-12的相关文章,限定文章语言种类为中文,检索词“缺氧诱导因子”。资料选择:对资料进行初审,纳入标准:①缺氧诱导因子介绍。②缺氧诱导因子对糖酵解的影响。资料提炼:共收集到符合上述要求的文献23篇,排除8篇重复性研究,15篇符合纳入标准。资料综合:缺氧诱导因子是一个基本的循环转录因子,当哺乳动物细胞在缺氧、促红细胞生成素和血管内皮生长因子或一些蛋白基因转录激活时被表达。它在维持氧稳态时起着非常重要的作用。当前的研究也为缺氧诱导因子在调节一些糖酵解酶的基因编码提供了证据,缺氧诱导因子作为一个重要的信号转导途径,糖酵解酶的基因启动子作为低氧反应元件和缺氧诱导因子结合并受缺氧诱导因子激活。结论:缺氧诱导因子在机体处于低氧状态时对糖酵解的激活起着非常重要的作用。

【Abstract】 OBJECTIVE:To summarize and analyze the main metabolic pathway of hypoxia-inducible factor and its regulation on glycolytic enzyme,explore the potential mechanisms of long time and high workload training or highland training in order to relieve the harm of strenuous exercise on body.DATA SOURCES:The relevant articles to related hypoxia-inducible factor published between January 1993 and December 2004 were searched for in Medline with the Keywords of "Hypoxia-inducible factor 1".Similarly,the China Journal Full-text Database was searched for related articles published between January 1993 and December 2004 with the key words of "Hypoxia-inducible factor" in Chinese.STUDY SELECTION:The literatures were selected primarily.The inclusive criteria were ①the introduction of hypoxia-inducible factor and ②the effect of hypoxia-inducible factor 1 on glycolytic enzymes.DATA EXTRACTION:There were 23 literatures in accordance with the above-mentioned criteria,and 8 of them were excluded due to the repetition of the same research,so 15 were selected.DATA SYNTHESIS:Hypoxia-inducible factor 1 was a basic helix-loop-helix transcription factor,which was expressed when mammalian cells were subjected to hypoxia and when transcription of genes encoding erythropoietin,vascular endothelial growth factor or some protein genes were activated.It was important for maintaining oxygen homeostasis.Previous studies had provided evidence that hypoxia-inducible factor 1 also regulated transcription of some genes encoding glycolytic enzymes.As a important signal transduction pathway,hypoxia-inducible factor 1 combined with promoters of glycolytic enzymes as a hypoxia response elements,while the promoters of glycolytic enzymes was activated by hypoxia-inducible factor 1.CONCLUSION:Hypoxia-inducible factor 1 plays an essential role in activating glycolytic enzymes in hypoxia condition.
