滋补肝肾中药对大脑中动脉闭塞大鼠脑勿动蛋白A表达的影响

【摘要】 目的:观察滋补肝肾中药对大脑中动脉闭塞模型大鼠脑内勿动蛋白A的影响,探讨中药促进神经发生的作用途径。方法:实验于2004-09/2005-01在山东省医学分子生物学重点实验室完成。30只大鼠随机分为药物组、模型组、假手术组,每组10只。用Tamura法制备大脑中动脉闭塞大鼠模型,于造模成功后6天药物组给予中药复方复健片水溶液10g/kg灌胃,复健片主要成份为何首乌、草决明、桑寄生、海马、淫羊藿等,由山东鲁信药业有限公司生产。其余两组分别灌胃给予同等量生理盐水2周,1次/天。原位杂交法观察大脑中动脉闭塞大鼠脑内勿动蛋白-A的表达,并对切片进行图像分析,测定染色平均灰度。结果:30只Wistar大鼠纳入结果分析。模型组给药1周后大鼠脑内勿动蛋白染色平均灰度为122.60±10.96,药物组为104.07±14.00,两组比较差异有显著性(P<0.01);2周后模型组大鼠脑内勿动蛋白染色平均灰度为132.48±7.77,药物组为98.09±10.33,两组比较差异有显著性意义(P<0.01)。同组1周与2周比较差异均无显著性(P均>0.05)。假手术组大鼠脑内勿动蛋白染色平均灰度为130.96±1.23,与药物组比较差异显著(P<0.01),与模型组比较差异无显著性(P>0.05)。结论:中药复方复健片可显著抑制大脑中动脉闭塞大鼠脑内勿动蛋白A的表达,从而促进缺血性卒中神经发生。

【Abstract】 AIM: To study the effect of Chinese herb for tonifying liver and kidney on the expression of Nogo protein-A in the brain of rats with middle cerebral artery occlusion (MCAO) and explore its action mechanism to promote neurogenesis. METHODS: The experiment was conducted in the Key Laboratory of molecular biology of Shandong province between September 2004 and January 2005. Totally 30 rats were randomly divided into drug group, model group and sham-operation group with 10 rats in each group. The rat models of MCAO were successfully established by the Tamura method. Six days later, the rats of the drug group were given oral 10 g/kg Chinese herb of fujian tablets solution (composed of heshouwu, caojueming, sangjisheng, haima, yinyanghuo and so on, and produced by Shandong Luxin Pharmaceutical Co., Ltd). The rest groups were given oral normal saline at the same dose once a day for two weeks. The expression of Nogo protein-A in the brain of MCAO rats was observed by in situ hybridization and the section was subjected to image analysis to determine the mean staining gray. RESULTS: All the 30 rats were involved in the result analysis. After one week of administration, the mean gray value of the expression of Nogo protein-A in the model group was 122.60±10.96, the drug group 104.07±14.00, which had significant differences (P < 0.01); 2 weeks later, the value in the model group was 132.48±7.77, the drug group was 98.09±10.33, obviously decreased which had significant differences (P < 0.01). There was no significant difference between the results after 1 week and 2 weeks in the same group (all P > 0.05). the mean gray value of the sham-operation group was 130.96±1.23, which had significant differences compared with the drug group (P < 0.01), but no significant difference compared with the model group (P > 0.05). CONCLUSION: Chinese herb of fujian tablets can predominantly inhibit the expression of Nogo protein-A in the brain of MCAO rats, so as to promote the neurogenesis of acute ischemia cerebral disease.
