穹隆海马伞切断与卵巢切除大鼠皮质区及Meynert核区雌激素受体α的表达

【摘要】 目的:观察穹隆海马伞切断与卵巢切除大鼠脑内皮质区、基底前脑Meynert核区雌激素受体α表达的变化,探讨胆碱能和雌激素系统与认知障碍相关的发病过程。方法:实验于2003-03/12在青岛大学医学院附属医院脑病防治重点实验室(省级重点实验室)完成。①分组:成年健康雌性Wistar大鼠28只,分为穹隆-海马伞切断组、卵巢切除组、穹隆-海马伞切断加卵巢切除组、对照组,每组7只。②模型制备:穹隆-海马伞切断组动物麻醉固定于脑立体定位仪后,于前囟后2.2~2.5mm,中线外1mm处凿开颅骨,切开硬脑膜,自制双刃刀完全切断海马伞外侧缘。双侧卵巢切除组动物麻醉后,打开腹腔,在肾脏下方的脂肪内取出卵巢,结扎并切除。对照组除不切断卵巢和穹隆-海马伞外,其余步骤同模型组。③免疫细胞化学染色法观察皮质区和基底前脑Meynert核区雌激素受体α阳性细胞表达变化。Olympus光学显微镜下(10×40倍)计数平均单个视野雌激素受体α阳性细胞的数量。结果:28只大鼠均进入结果分析。在皮质区,Meynert核区雌激素受体α表达阳性细胞数:双侧穹隆-海马伞切断组,双侧卵巢切除组,双侧穹隆-海马伞切断加双侧卵巢切除组均明显低于对照组[皮质区:(25.71±1.11),(18.29±7.99),(20.31±8.96),(58.69±30.26)个/视野;Meynert核区:(3.31±1.11),(3.83±1.34),(2.26±2.45),(52.31±22.52)个/视野,(P<0.01)]。结论:穹隆-海马伞切断和/或卵巢切除可以影响大鼠皮质区和Meynert核区雌激素受体α的表达,认为胆碱能系统与雌激素系统可能存在内在的相互作用的病理过程。

【Abstract】 AIM: To discuss expression of estrogen receptor-α (ER-α) in cerebral cortex and Meynert nucleus in brain of rats with fimbria/fornix transection and ovariectomy, and study the pathologic process between the cholinergic system and the estrogen system. METHODS: The experiment was completed in the Institute of Cerebrovascular Disease, Affiliated Hospital of Qingdao University Medical College from March to December 2003. ① Grouping: Twenty-eight healthy female rats were randomly divided into four groups: ovariectomy group, fimbria/fornix transection group, fimbria/fornix transection + ovariectomy group and control group with 7 in each group. ② Model establishing: After anaesthesia, rats in fimbria/fornix transection group were fixed on brain solid positioner; skull was cut off at the bregma anterior-posterior 2.2 mm-2.5 mm and lateral 1.0 mm; dura was cut off and lateral border of fimbria of hippocampus was cut off completed with a double blade. After anaesthesia, abdomen of rats in bilateral ovariectomy group was opened, and the bilateral ovaries in the fattiness under the kidney were ligated and ablated. Rats in control group were treated with the same ways as compared with those in model group except fimbria/fornix transection and ovariectomy. ③ Changes of ER-α positive expression were observed with immunocytochemical stain in cortex and Meynert nucleus. The amounts of the ER-α positive neurons were observed under the Olympus microscope (10×40). RESULTS: All 28 rats entered the final analysis. Positive cellular number of ER-α in cerebral cortex and Meynert nucleus: Numbers in bilateral fimbria/fornix transection group, bilateral ovariectomy group and bilateral fimbria/fornix transection + bilateral ovariectomy group were lower than those in control group Cortical area: (25.71±1.11), (18.29±7.99), (20.31±8.96), (58.69±30.26)/sight; Meynert nucleus: (3.31±1.11), (3.83±1.34), (2.26±2.45), (52.31±22.52)/sight, P < 0.01. CONCLUSION: Fimbria/fornix transection and/or ovariectomy can affect the expression of ER-α in the cortex and Meynert nucleus, and a pathologic relationship between the cholinergic system and the estrogen system may exist under this condition.
