IL-1β或IL-6致痫对大鼠大脑皮质一氧化氮合酶表达的影响
Effect of IL-1β or IL-6 Induced Epilepsy on the Expression of NOS in Cerebral Cortex of Rats
【摘要】 目的探讨白细胞介素-1β(IL-1β)和白细胞介素-6(IL-6)致痫对大鼠大脑皮质一氧化氮合酶(NOS)表达的影响。方法采用免疫印迹技术和图像分析技术,实验大鼠行侧脑室注射IL-1β或IL-6及经糖皮质激素(GC)预处理后再行侧脑室注射IL-1β或IL-6,观察其行为学的改变,检测脑内NOS表达的变化。结果动物行为学观察显示IL-1β组、IL-6组癫痫发作程度达中度,GC+IL-1β组、GC+IL-6组和单纯GC组则无明显癫痫发作。IL-1β组I、L-6组大脑皮质NOS含量明显高于对照组(均P<0.05),GC+IL-1β组、GC+IL-6组和单纯GC组与对照组相比,差别均无显著性意义(均P>0.05)。结论IL-1β或IL-6致痫机制与脑内一氧化氮信号转导通路有关,GC具有明显抑制NOS表达的作用和抗痫效应。
【Abstract】 Objective To explore the effect of IL-1β or IL-6 on the expression of NOS in cerebral cortex of epileptic rats.Methods IL-1β or IL-6 was injected into the lateral cerebral ventricle or following pretreatment with glucocorticoids in the experimental rats.The behavioral changes were observed.The content of NOS in cerebral cortex was detected by using Western blot and image analysis technique.Results Observation on the animal behaviors revealed that the extent of epileptic attack was moderate in IL-1β and IL-6 groups.There was no epileptic attack in control group,GC +IL-1β group,GC +IL-6 group and GC alone group.The content of NOS in cerebral cortex was significantly increased in IL-1β group and IL-6 group(P<(0.05)) as compared with that in the control group,while there was no significant difference among GC +IL-1β group,GC+IL-6 group and GC alone group(P>0.05) and control group.Conclusion The epilepsy induced by IL-1β or IL-6 may relate to the signal transduction pathway of NO.Glucocorticoids can inhibit the expression of NOS and suppress the epilepsy attack.
