N-苄基氟哌啶醇氯化物对大鼠血管平滑肌细胞内钙离子的影响

【摘要】 目的:动态观察N基氟哌啶醇氯化物(F3)对大鼠胸主动脉平滑肌细胞内Ca2+的影响。方法:用Ca2+荧光染料Fluo3AM负载大鼠胸主动脉平滑肌细胞,在激光扫描共聚焦显微镜上测定细胞内游离钙的变化。结果:在含CaCl2(1.26mmol/L)的细胞外液中,30 mmol/L KCl使细胞内钙荧光强度(FI)迅速增强,F3可以拮抗KCl诱导钙FI增强作用,并呈量效依赖和时间依赖关系。4种浓度F3(0.01、0.1、1.0、10μmol/L)作用3 min时,使KCl诱导细胞内钙FI从(81±4)%分别降至(71±18)%(n=20,P<0.05)、(51±12)%(n=20,P<0.01)、(39±14)%(n=20,P<0.01)、(28±14)%(n=20,P<0.01)。钙FI变化最快的时程是在给药后的030 s。结论:F3通过阻断细胞膜电压依赖性钙通道降低平滑肌细胞内钙浓度,这可能是其扩张血管,改善心肌缺血的机制。

【Abstract】 Objective:) To investigate the effects of N-benzyl haloperidol(chloride(F-3)on) intracellular calcium([Ca2+]-i)in vascular smooth muscle cells(VSMCs)isolated from thoracic aortas of rat.Methods: VSMCs were loaded with a calcium sensitive fluorescent dye Fluo-3-AM,and [Ca2+]-i was determined by the use of laser scanning confocal microscope.Results: In the extracellular solution with Ca2+ at the concentration of 1.26 mmol/L,30 mmol/L KCl induced a rapid increase of intracellular calcium fluorescence intensity(FI).F-3 antagonized the increase of FI induced by KCl in both concentration dependent manner and time dependent manner.After 3 minutes of the administration of F-3 at the concentration of 0.01,0.1,1.0,10 μmol/L,the increased FI of the VSMCs induced by KCl was decreased from(81±4)% to(71±18)%(n=20,P<0.05),(51±12)%(n=20,P<0.01),(39±14)%(n=20,P<0.01),(28±14)%(n=20,P<0.01),respectively.During 0~30 seconds after the administration of drugs,the changes of FI were more ra-pid.Conclusion: F-3 reduces[Ca2+]-i via inhibition of voltage dependent calcium channel,which is possibly involved in the mechanism of its vasodilatory and anti-myocardial ischemic effects.
