响应面法分析效应物对苯丙氨酸解氨酶活性的影响

【摘要】 实验考察了聚乙二醇6000、D-山梨醇、海藻糖、二巯基苏糖醇、甘油、戊二醛对酶反应中苯丙氨酸解氨酶活力的影响。单因子实验发现,0.5%聚乙二醇6000对PAL酶活的影响最大,可提高活力80.3%。响应面实验发现,海藻糖、聚乙二醇6000、二巯基苏糖醇在实验浓度范围内交互作用很小,0.62%海藻糖、0.54%聚乙二醇6000和0.31%巯基苏糖醇的组合可提高酶活92.9%。

【Abstract】 The effects of six effectors, polyethylene glycol (PEG) 6000, D-sorbitol, trehalose, dithiothreitol, glycerol and glutarie dia|dehyd on PAL activity in enzymatic reaction were investigated in this article. By single factor experiment, 0.5% PEG was found to significantly improve PAL activity by 80.3%. By response surface experiment, it was found that the combination use of 0.54% PEG6000, 0.62% trehalose and 0.31% DTT could effectively improve the PAL activity by 92.9%.
