【摘要】 目的观察补肾益精中药对体外培养成骨细胞类胰岛素样生长因子I(IGF-I)分泌和表达的影响,探讨补肾益精中药调节骨重建的作用机理。方法采用连续酶消化法培养大鼠颅盖骨成骨细胞,观察补肾益精药物血清对成骨细胞增殖率、碱性磷酸酶(ALP)、矿化结节数目以及成骨细胞不同分化时期IGF-I分泌和mRNA表达的影响。结果与对照组比较,补肾益精中药能明显增加成骨细胞增殖率,提高成骨细胞ALP分泌量,增加矿化结节数;中药组IGF-I的分泌在第7d、第21d时与对照组比较无差异,但在第14d时IGF-I的分泌量明显高于对照组,同时其成骨细胞IGF-I mRNA表达下调。结论补肾益精中药可能通过调控细胞因子IGF-I的表达与分泌发挥促进骨形成的作用。更多还原

【Abstract】 Objective To study the effects of kidney-nourishing and essence-enriching herbs on osteoblast IGF-I secretion and expression and its treatment mechanism on osteoporosis.Methods Continuous enzyme digestion method was used to cultivate rat skull osteoblast and further observe the osteoblast proliferation rate,ALP,numbers of mineralized nodules,and IGF-I secretion and mRNA expression in various differential stages.Results Compared with the control group,these herbs could obviously increase osteoblast proliferation rate and ALP secretion,and the numbers of mineralized nodules;on the seventh and twenty-first days,IGF-I secretion was similar in both control and treatment groups;but on the fourteenth day,IGF-I secretion was higher in the treatment group than that of the control group,and the osteoblast IGF-I mRNA expression was down-regulated.Conclusion Kidney-nourishing and essence-enriching herbs can promote bone formation by regulating IGF-I expression.更多还原
