抗HER-2嵌合抗体chA21对人卵巢癌SKOV3细胞凋亡的诱导作用及其机制

【摘要】 目的探讨抗HER-2嵌合抗体chA 21在体外对高表达HER-2的人卵巢癌SKOV 3细胞的凋亡诱导作用及其分子机制。方法chA 21处理体外培养的SKOV 3细胞,采用透射电镜和原位末端标记技术观察和检测细胞凋亡情况;采用免疫细胞化学技术检测凋亡相关基因B cl-2、B ax、F as及天冬氨酸特异性半胱氨酸蛋白酶(caspase-3)表达情况。结果chA 21作用72h,可见SKOV 3细胞凋亡,其作用呈剂量依赖性;SKOV 3细胞B ax、F as和caspase-3表达升高、B cl-2表达下调、B cl-2/B ax降低,并呈剂量依赖性。结论chA 21在体外可诱导SKOV 3细胞凋亡,其分子机制与调节凋亡相关基因B ax、B cl-2、F as及caspase-3的表达有关。

【Abstract】 Objective: To explore apoptosis of SKOV3 cells induced by anti-HER-2(chimeric) antibody chA21 in vitro and its possible mechanisms.Methods: Apoptosis of SKOV3 cells induced by chA21 was examined by electron microscopy and TUNEL staining.The expression of Bax,Bcl-2,Fas and caspase-3 in SKOV3 cells after being exposed to chA21 were measured with immunocytochemistry.Results: Apoptotic index of SKOV3 cells increased dose-dependently after being treated with chA21;Expression of Bax,Fas,caspase-3 increased and Bcl-2 decreased,Bcl-2/Bax ratio decreased dose-dependently.Conclusions: The results indicate that chA21 can remarkably induce apoptosis of SKOV3 cells by regulating the expression of Bax,Bcl-2,Fas and caspase-3.
