HIVgp120对胎鼠背根神经节神经元突起生长的影响
The effects of HIVgp120 on cultured dorsal root ganglion neuronal growth
【摘要】 目的:探讨人类免疫缺陷病毒(human immunodeficiency virus,HIV)gp120对体外培养的背根神经节神经元胞体和突起生长的影响。方法:分散培养的胎鼠背根神经节神经元用不同浓度的HIVgp120(125、250、500 pmol/L和1 nmol/L)处理,对分散培养的背根神经节神经元用MAP2免疫荧光标记后,用共聚焦激光扫描显微镜观察神经元胞体和突起的改变。结果:应用gp120后7 d,神经元突起的数目减少,长度变短,而神经元的胞体则没有明显变化。结论:HIV gp120可直接影响分散培养的背根神经节神经元突起的生长。
【Abstract】 Objective: To determine the effects of human immunodeficiency virus(HIV) gp120 on dissociated cultured dorsal root ganglion(DRG) neuronal growth.Methods: Dissociated cultured rat embryo′s DRG cells were treated with HIV gp120 at different concentration(125?pmol/L,250?pmol/L,500?pmol/L,1?nmol/L,respectively) and then were processed for MAP2 immunofluorescent labeling,and furthermore the changes of neuronal cell body and neurites were observed with confocal laser scanning microscope.Results: The number and length of neurites were reduced after 7day treatment with gp120 compared with control group,whereas the diameter of neuronal cell body was not obviously altered compared with control group.Conclusion: HIV gp120 may have direct influences on the outgrowth of neurites in dissociated DRG cell cultures.
