【摘要】 目的建立人血清催乳素的时间分辨荧光免疫分析(TRFIA)方法。方法采用双抗体夹心法建立人血清催乳素TRFIA检测试剂盒,对试剂盒的各项指标进行评价。结果试剂盒的定量范围为16～11000mU/L,灵敏度为1.36mU/L,批内批间的精密度分别为2.6%～7.6%,5.7%～9.0%;与人胎盘泌乳素(hPL)、人生长激素(hGH)、人黄体生成素(hLH)和人促卵泡激素(hFSH)无交叉反应。30份异常血样用本试剂盒与进口的同类试剂盒同时检测,其相关系数为0.997。结论试剂盒各项指标均达到临床检测要求,可替代国外同类产品试剂盒。更多还原

【Abstract】 Objective To prepare a time-resolved fluoroimmunoassay (TRFIA) kit for detection of human prolactin. Method A sandwich TRFIA kit for detection of human prolactin was developed using monoclonal antibody of anti-prolactin. Result The measure range of the kit was 16～11000 mU/L. Its assay sensitivity was 1.36 mU/L. There was no cross reactivity with human placental lactogen, human growth hormone, human luteinizing and human follicle stimulating hormones. The intra- and inter-assay coefficients of variation were 2.6%～7.6% and 5.7%～9.0%, respectively. The correlation coefficient of blood samples detection results between home-made and commercially available PRL-DELFIA (PerKin Elmer) was 0.997. Conclusion The developed prolactin TRFIA kit in this study was a valuable test for clinical application with better sensitivity, specificity, and accuracy.更多还原
