小鼠肝癌微波消融对宿主Th2/Th1免疫应答的影响
Microwave Ablation of Mouse Hepatocellular Carcinoma:Effects on the Th2/Th1 Immune Response in the Host
【摘要】 目的探讨小鼠肝癌微波消融(microwaveablation,MA)对宿主Th2/Th1免疫应答的影响。方法建立C57BL/6J小鼠Hepa1-6细胞移植肝癌模型,对肿瘤微波消融或手术切除,取正常、荷瘤、手术后及MA后四组小鼠,每组6只,用ELISA法检测各组周围血血浆Th1(IL-12I、FN-γ)和Th2(IL-4I、L-10)细胞因子水平(pg/ml)。结果荷瘤组比正常组,细胞因子IL-10增加,IL-4I、L-12和IFN-γ下降(均P>0.05)。MA组较荷瘤组,IL-12、IFN-γ显著上升(均P<0.01),IL-4明显降低(P<0.05),IL-10有所下降(P>0.05)。手术组比荷瘤组,IL-12I、FN-γ上升,IL-4I、L-10下降(均P>0.05)。MA组比手术组,IL-12明显上升(P<0.05),IFN-γ升高,IL-4和IL-10下降(均P>0.05)。结论MA原位灭活小鼠肝癌可促进宿主免疫应答的Th2/Th1偏移,并且MA较手术切除具有较强的免疫刺激效应。
【Abstract】 Objective To investigate the effect of microwave ablation(MA) of hepatocellular carcinoma(HCC)on the Th2/Th1 immune response in tumor-bearing mice.Methods A HCC model was established by subcutaneous inoculation of syngeneic Hepa1-6 cells into C57BL/6J mice.The tumors were treated with MA or by resection.Four groups of animals(6 mice per group),namely normal,tumor-bearing,resection-treated and MA-treated,were set up.Concentrations of Th1(IL-12,IFN-γ) and Th2(IL-4,IL-10) cytokines in plasma were determined by ELISA.Results Compared with the normal mice,the levels of IL-4,IL-12 and IFN-γ in tumour-bearing mice were decreased while the level of IL-10 was increased(P >0.05).For the comparisons between MA and tumor-bearing group,only the levels of IL-12 and IFN-γ in the MA group were markedly higher than the tumour-bearing mice(P<0.01).Compared with tumor-bearing mice,the levels of IL-12 and IFN-γ in mice treated by resection were increased,but the levels of IL-4 and IL-10 were lowered(P>0.05).The level of IL-12 in MA group was obviously higher than the mice with tumour resection(P<0.05).With the exception of IL-10 and IL-4,the level of IFN-γ in mice treated with MA was higher than the mice with tumour resection(P>0.05).Conclusion A skewing of Th2/Th1 immune response was seen in mice after MA of HCC.MA of the tumor may cause a stronger immune reaction than resection.
