酵母菌SDU2.91中菊粉酶的研究
Studies on the Inulinase of Yeast Strain SDU2.91
【摘要】 研究了隐球酵母菌SDU2.91的产菊粉酶条件,优化的产酶培养基为:菊粉2%、牛肉浸膏0.5%、(NH4)2HPO40.5%、MgSO4.7H2O 5mmol/L,pH 5~6。发酵32h后,发酵液酶活达到最高,为28u/m l。所产的菊粉酶主要定位于细胞壁,胞外酶胞壁酶胞内酶比为10 85 5。酶最适温度为55℃,最适合pH为5.0。其I/S的比值为0.96,酶解产物以果糖为主。
【Abstract】 The conditions for producing inulinases by the strain of Cryptococcus sp.SDU 2.91 were studied.The results indicated that the optimum conditions were inulin 2%,beef extract 0.5%,(NH4) 2PO4 0.5%,MgSO4·7H2O 5mmol/L,pH 5-6.The inulinase activity was 28u/ml when the strain cultured at above conditions for 32h.The distribution of inulinase was discussed and the ratio of inulinase from extra-cell,cell-wall,intra-cell was 10855.The optimal temperature and pH of the enzyme was 55℃ and 5.0 respectively.The I/S of enzyme is 0.96.The enzymolysis products were mainly fructose.
